为建立测定血浆中甲基强的松龙(MP)及其前体酯甲基强的松龙琥珀酸酯(MPHS)的高效液相色谱(HPLC)法。采用HPLC法检测大鼠静脉注射甲基强的松龙琥珀酸钠(MPS)后不同时间血浆中MP及MPHS的浓度。采用YWG—C_18分析柱,以乙腈∶水∶醋酸—醋酸钠缓冲液(29∶71∶1.4,V/V/V)为流动相,在254nm波长下监测洗提液。结果标准曲线的相关系数大于0.9993,日内及日间变异系数分别为(1.9~5.6)%和(4.1~14.7)%。结果提示本法系国内首创,具有操作简便、快速、灵敏、准确等特点。
To establish a high performance liquid chromatography (HPLC) method for the determination of methylprednisolone succinate (MPHS) in plasma and its precursor methylprednisolone (MP). The concentrations of MP and MPHS in plasma at different time points after intravenous injection of methylprednisolone sodium succinate (MPS) in rats were determined by HPLC. The eluate was monitored at 254 nm using a YWG-C 18 analytical column with acetonitrile: water: acetic acid-sodium acetate buffer (29:71: 1.4, V / V / V) as the mobile phase. The correlation coefficient of the standard curve was greater than 0.9993. The intra-day and inter-day coefficient of variation were (1.9-5.6)% and (4.1-14.7)%, respectively. The results suggest that this law is the first domestic initiative, with simple, fast, sensitive and accurate.
