目的:观察决明子蒽醌苷与山楂总三萜酸配伍对实验性高血脂症兔血清低密度脂蛋白胆固醇(low densit ylipoprotein-cholesterol LDL-C)及肝细胞膜高密度脂蛋白受体(high density lipoprotein receptor HDLR)活性的影响。方法:雄性新西兰兔高脂饲养2周形成实验性高血脂症,继续给予高脂饮食,然后采用均匀设计法,分别给予决明子和山楂有效部位不同的比例配伍灌服,4周后高脂给量减半,继续服药至6周,免疫比浊法测定血清LDL-C变化,以125I标记的兔HDL3作为配体,用放射受体分析法对肝细胞膜HDLR活性进行测定。结果:决明子蒽醌苷与山楂总三萜酸按1∶2.63配伍,可使血清LDL-C含量明显降低(P<0.01),肝细胞膜HDLR活性增加(P<0.05)。结论:决明子蒽醌苷与山楂总三萜酸最佳配比降低高血脂兔血清LDL-C的作用明显强于单一组分及原药对配伍,对肝细胞膜HDLR的影响呈现以受体数目显著增加为特征的受体结合活性升高。
OBJECTIVE: To observe the effects of combination of cassia seed anthraquinone glycosides and hawthorn total triterpene acid on the serum levels of serum low density lipoprotein cholesterol (LDL-C) and high density lipoprotein (HDL-C) in experimental hyperlipidemia rabbits receptor HDLR activity. METHODS: Male New Zealand white rabbits were fed with high fat diet for 2 weeks to develop experimental hyperlipidemia. The high fat diet was continued. Then uniform design method was used to give the different proportion of effective parts of cassia seed and hawthorn. Halved and continued to medication until 6 weeks, serum LDL-C changes by turbidimetric assay, 125L labeled rabbit HDL3 as a ligand, determination of HDLR activity of liver cell membrane by radioimmunoassay. Results: The combination of cassia anthraquinone glycoside and hawthorn total triterpene acid at 1:2.63 could significantly decrease the serum LDL-C content (P <0.01) and increase the HDLR activity of hepatocyte membrane (P <0.05). CONCLUSION: The optimal ratio of anthraquinone glycoside and hawthorn triterpene in the cassia seed reduces the effect of serum LDL-C in hyperlipidemic rabbits significantly stronger than that of single component and original drug pairs.
