目的:研究麝香、冰片、薯蓣皂苷及栀子苷对大鼠脑缺血再灌注炎性损伤的保护作用。方法:将165只大鼠分为假手术组、模型对照组、麝香、冰片、薯蓣皂苷及栀子苷大小剂量组,醒脑静组。各组预防给药4d后,采用线栓法复制大鼠脑缺血再灌注模型,缺血24小时后,考察麝香、冰片、薯蓣皂苷及栀子苷对缺血再灌注后对神经功能损伤的改善作用,脑组织环氧化物酶(COX-2)及5脂氧酶(5-LOX)活性的影响。结果:麝香50mg/kg组及冰片50mg/kg栀子苷30mg/kg组能明显改善神经损伤引起的行为学异常;麝香组、冰片25mg/kg组、薯蓣皂苷60mg/kg组及栀子苷60mg/kg组可降低缺血再灌注后脑内COX-2的活力;麝香组、冰片50mg/kg组及薯蓣皂苷、栀子苷60mg/kg组大鼠脑组织可明显降低缺血再灌注后脑内5-LOX活性。结论:麝香、冰片、薯蓣皂苷及栀子苷对脑缺血再灌注急性期炎性损伤有一定的保护作用。
Objective: To study the protective effects of musk, borneol, diosgenin and geniposide against inflammatory injury induced by cerebral ischemia-reperfusion in rats. Methods: 165 rats were divided into sham operation group, model control group, musk, borneol, diosgenin and geniposide dose group and Xingnaojing group. After 4 days of administration, the rat model of cerebral ischemia-reperfusion was reproduced by thread occlusion method. After 24 hours of ischemia, effects of musk, borneol, diosgenin and geniposide on neurological damage after ischemia-reperfusion (COX-2) and 5-lipoxygenase (5-LOX) activity in brain tissue. Results: Musk group, borneol 25mg / kg group, diosgenin 60mg / kg group and jasminoidin 60mg / kg group could significantly improve the behavioral abnormalities caused by nerve injury in musk 50mg / kg group and borneol 50mg / kg geniposide 30mg / / kg group could reduce the activity of COX-2 in the brain after ischemia-reperfusion; The brain tissue in the musk group, borneol 50mg / kg group and diosgenin, geniposide 60mg / kg group could obviously reduce the content of 5 -LOX activity. Conclusion: Musk, borneol, diosgenin and geniposide have some protective effects on the inflammatory injury induced by acute cerebral ischemia and reperfusion.
