目的 :在实验室通过病毒分离和传播试验 ,确定致倦库蚊是否能传播登革病毒。方法 :1.用大剂量登革Ⅱ型病毒 (病毒滴度为 10 - 9~ 10 - 10 TCID50 )同时喂食致倦库蚊和白纹伊蚊 ,不同时间后用C6/ 36细胞从蚊体内分离登革病毒。 2 .用吸食病毒 7d后的致倦库蚊和白纹伊蚊攻击小白鼠 1d龄乳鼠 ,两周后取鼠血清进行免疫荧光试验。结果 :13d内不同时间均能从白纹伊蚊体内分离出登革病毒 ,而致倦库蚊在 4 8h以后不能分离到登革病毒 ,表明登革病毒在致倦库蚊体内已经消失 ;白纹伊蚊攻击组的小白鼠血清中登革Ⅱ型病毒抗体为阳性 ,而致倦库蚊攻击组的小白鼠血清中的抗体为阴性 ,说明致倦库蚊并不能将登革病毒通过刺叮传播给敏感的乳鼠。结论 :致倦库蚊不能作为登革病毒的传播媒介
OBJECTIVE: To determine whether Culex pipiens quinquefasciatus can transmit dengue virus through virus isolation and transmission tests in laboratories. Methods: 1. Culex pipiens quinquefasciatus and Aedes albopictus were fed with high dose dengue virus type Ⅱ (virus titer of 10 - 9-10-10 TCID50), and were separated from the mosquito by C6 / 36 cells after different time Dengue virus. The mice were challenged with Culex pipiens quinquefasciatus and Aedes albopictus for 7d after ingestion of virus for 1d, and serum of mice was taken two weeks later for immunofluorescence assay. Results: Dengue virus was isolated from Aedes albopictus at different times within 13d, while dengue virus could not be isolated after 48h, indicating that dengue virus had disappeared in Culex quinquefasciatus. White Aedes mosquitoes attack the group of mice serum dengue type II antibody was positive, and Culex pipiens quinquefasciatus attack in mice serum antibody was negative, that Culex pipiens pallens can not be infected with dengue virus through thorns Spread to sensitive suckling mice. Conclusion: Culex pipiens quinquefasciatus can not serve as a transmission vector of dengue virus
