目的:研究玻璃体腔注入热休克蛋白27(HSP27)对大鼠慢性青光眼模型血清中HSP27抗体浓度的影响。方法:51只Wistar大鼠随机分为高眼压组(15只),假手术对照组(15只),致敏组(9只),致敏对照组(9只),正常组(3只),右眼为实验眼,左眼为对照眼。术前、术后第3天、1周、2周、4周、6周、8周均检测大鼠双眼眼压。按时间点依次处死大鼠,抽血供酶联免疫吸附(ELISA)法检测HSP27抗体含量。结果:①高眼压组、致敏组和致敏对照组右眼术后眼压明显升高(p<0.01);②高眼压组和致敏对照组术后血清中HSP27抗体浓度明显升高(P<0.05),致敏组术后血清HSP27抗体浓度未见升高(p>0.05)。结论:①随着眼压升高和持续时间延长,大鼠血清中的HSP27抗体水平逐渐升高;②玻璃体腔内注射HSP27能完全抑制高眼压状态下血清中HSP27抗体水平的升高。
Objective: To investigate the effect of intravitreal injection of heat shock protein 27 (HSP27) on the concentration of HSP27 in the serum of chronic glaucoma model rats. Methods: Fifty-one Wistar rats were randomly divided into high intraocular pressure (IOP) group (15), sham operation control group (15), sensitized group (9), sensitized control group (9), normal group ), The right eye is the experimental eye, the left eye is the control eye. Preoperative, postoperative day 3, 1 week, 2 weeks, 4 weeks, 6 weeks, 8 weeks were measured intraocular pressure in rats. Rats were sacrificed at different time points and blood was collected for enzyme-linked immunosorbent assay (ELISA) to detect the content of HSP27 antibody. Results: ① The intraocular pressure of the patients with ocular hypertension, sensitized group and sensitized control group were significantly increased (p <0.01); ② The serum concentration of HSP27 in the ocular hypertension group and the sensitized control group were significantly increased (P <0.05). The serum HSP27 antibody concentration in sensitized group was not increased (p> 0.05). Conclusions: ①With the increase of intraocular pressure and prolongation of duration, the level of HSP27 antibody in serum of rats gradually increased; ② The intravitreal injection of HSP27 could completely inhibit the elevation of HSP27 in serum under ocular hypertension.
