目的:检测RORγt基因在西宁地区免疫性血小板减少症(ITP)外周血单个核细胞中的表达及其临床意义。方法:应用实时荧光定量PCR对30例西宁地区免疫性血小板减少症患者及20例正常对照者外周血单个核细胞中RORγt mRNA进行检测。结果:ITP组(治疗前:0.958±0.264;治疗后:0.604±0.151)RORγt基因mRNA表达水平较对照组(0.511±0.160)升高,ITP组治疗后RORγt基因mRNA表达水平下降。治疗前后ITP患者RORγt水平与外周血血小板计数存在负相关(r=-0.739,P<0.05 and r=-0.687,P<0.05)。结论:RORγt在西宁地区ITP患者外周血单个核细胞中高表达,可能参与ITP的发生发展,可能有助于ITP的早期诊断、治疗及预后。
Objective: To detect the expression of RORγt gene in peripheral blood mononuclear cells of immune-thrombocytopenia (ITP) in Xining and its clinical significance. Methods: RORγt mRNA was detected in peripheral blood mononuclear cells of 30 patients with immune thrombocytopenia and 20 normal controls by real-time fluorescence quantitative PCR. Results: The mRNA expression of RORγt in ITP group (before treatment: 0.958 ± 0.264; after treatment: 0.604 ± 0.151) was higher than that in control group (0.511 ± 0.160), and decreased in ITP group. There was a negative correlation between RORγt level and peripheral blood platelet count in ITP patients before and after treatment (r = -0.739, P <0.05 and r = -0.687, P <0.05). CONCLUSION: RORγt is highly expressed in peripheral blood mononuclear cells of ITP patients in Xining area, which may be involved in the development of ITP. It may contribute to the early diagnosis, treatment and prognosis of ITP.
