目的研究阻断氯通道对低分化鼻咽癌(CNE-2Z)细胞凋亡和细胞增殖的影响,探讨氯通道在CNE-2Z细胞凋亡中的作用。方法用顺铂(CDDP)诱导细胞凋亡,流式细胞仪检测细胞凋亡,MTT法检测细胞增殖。结果顺铂(1.5μg/ml)处理细胞48 h后可检测出凋亡峰,氯通道阻断剂NPPB(50μmol/L)阻抑顺铂诱导的细胞凋亡及细胞增殖抑制。结论氯通道参与了CNE-2Z细胞凋亡过程的调控。
Objective To study the effect of blocking chloride channel on apoptosis and cell proliferation in poorly differentiated nasopharyngeal carcinoma (CNE-2Z) and to explore the role of chloride channel in CNE-2Z cell apoptosis. Methods Cisplatin (CDDP) was used to induce apoptosis. Flow cytometry was used to detect apoptosis. Cell proliferation was detected by MTT assay. Results The apoptotic peak was detected at 48 h after treatment with cisplatin (1.5 μg / ml). Inhibition of cisplatin-induced apoptosis and cell proliferation were inhibited by the chloride channel blocker NPPB (50 μmol / L). Conclusion Chloride channels are involved in the regulation of apoptosis in CNE-2Z cells.
