目的 :探讨不同反应靶细胞中白细胞介素 6 (IL 6 )对信号转导子与转录激活子 1(STAT1)的激活。方法 :利用抗STAT1和抗酪氨酸磷酸化STAT1的抗体 ,进行免疫印迹分析。结果 :IL 6可促进 7TD1和TF1细胞生长 ,但在这两种细胞中对STAT1的酪氨酸磷酸化水平没有明显影响。M 1、R2和U937细胞在IL 6诱导下生长停止并向巨噬细胞方向分化 ,而IL 6在这 3种细胞中均可激活STAT1,且激活效应有剂量与时间依赖性。结论 :STAT1的活化可能与抑制效应有关。我们的数据有助于解释为什么IL 6在不同效应靶细胞中有不同的生物学效应
AIM: To investigate the effects of interleukin-6 (IL-6) on signal transducers and activator of transcription 1 (STAT1) in different target cells. METHODS: Immunoblotting was performed using anti-STAT1 and anti-tyrosine phosphorylated STAT1 antibodies. Results: IL-6 promoted the growth of 7TD1 and TF1 cells, but had no significant effect on the level of tyrosine phosphorylation of STAT1 in both cells. M 1, R 2 and U 937 cells stopped their growth and induced macrophage differentiation under IL 6 induction. However, IL 6 activated STAT 1 in all three kinds of cells, and the activation effect was dose-dependent and time-dependent. Conclusion: The activation of STAT1 may be related to the inhibitory effect. Our data help explain why IL 6 has different biological effects in different effector target cells
