目的分析结核病患者外周血共刺激分子CD28/CTLA-4(CD152)表达,评价患者的免疫状态。方法采集21例初诊结核病患者及15例健康对照者外周血,采用多参数流式细胞术检测分析共刺激分子CD28、CD152的表达,以及分析其亚群CD4+CD152+细胞、CD4+CD28+细胞、CD8+CD28+细胞、CD8+CD152+水平的变化。结果与健康者比较,结核病患者外周血CD28+细胞、CD152+细胞水平明显增高(P<0.01)。对共刺激分子CD28、CD152进行亚群分析,结核病患者外周血CD4+CD152+细胞、CD4+CD28+细胞、CD8+CD152+细胞水平明显高于健康者(P<0.05),CD8+CD28+细胞水平无统计学意义(F=2.647,P=0.113)。结论结核病患者外周血共刺激分子CD28/CD152(CT LA-4)表达增加,且CT LA-4在CD4+和CD8+T细胞上表达均增加,提示可能存在T细胞活化或抑制信号调节失衡。
Objective To analyze the expression of costimulatory molecules CD28 / CTLA-4 (CD152) in peripheral blood of patients with tuberculosis and evaluate their immune status. Methods The peripheral blood samples from 21 newly diagnosed TB patients and 15 healthy controls were collected. The expression of costimulatory molecules CD28 and CD152 were analyzed by multiparameter flow cytometry. The subsets of CD4 + CD152 + cells, CD4 + CD28 + cells, CD8 + CD28 + cells, CD8 + CD152 + levels. Results Compared with healthy controls, the levels of CD28 + cells and CD152 + cells in peripheral blood of tuberculosis patients were significantly increased (P <0.01). The subgroups of CD28 and CD152 were analyzed by subgroup analysis. The levels of CD4 + CD152 +, CD4 + CD28 + and CD8 + CD152 + in peripheral blood of tuberculosis patients were significantly higher than those in healthy controls (P <0.05) Significance (F = 2.647, P = 0.113). Conclusion The expression of CD28 / CD152 (CT LA-4) in peripheral blood of patients with tuberculosis increased, and the expression of CT LA-4 in both CD4 + and CD8 + T cells increased, suggesting that T cell activation or inhibition might be unbalanced.
