以(4R-cis)-6-[2-[2-(4-氟苯基)-5-(1-异丙基)-3-苯基-4-[(苯胺)-羰基-1H-吡咯-1-基]乙基]-2,2-二甲基-1,3-二氧戊环-4-乙酸叔丁酯为起始原料,通过酸水解、碱水解,与醋酸钙中和3步得到目标化合物阿托伐他汀钙,研究了酸、溶剂和温度对反应的影响规律。优化反应条件为:水解采用盐酸,反应介质为乙醇,反应温度为55℃。在上述条件下,产品收率达88.7%。产物结构经核磁共振氢谱(1H NMR)和质谱(ESI-MS)确证。
(4-fluorophenyl) -5- (1-isopropyl) -3-phenyl-4 - [(aniline) -carbonyl-1H-pyrrole -1-yl] ethyl] -2,2-dimethyl-1,3-dioxolane-4-acetic acid tert-butyl ester as a starting material by acid hydrolysis, base hydrolysis and neutralization with calcium acetate 3 Step to obtain the target compound atorvastatin calcium, the acid, solvent and temperature on the reaction rule. The optimized reaction conditions were as follows: hydrochloric acid was used for hydrolysis and ethanol was used as the reaction medium. The reaction temperature was 55 ℃. Under the above conditions, the product yield reached 88.7%. The structure of the product was confirmed by 1H nuclear magnetic resonance (1H NMR) and mass spectrometry (ESI-MS).
