背景:利用一氧化氮的双重作用,找到治疗脑缺血的最佳药物、最佳给药时间和剂量等一直是研究的热点。目的:观察L-精氨酸和氨基胍对实验性大鼠局灶性脑缺血损伤的治疗作用以及一氧化氮在脑缺血损伤中的作用规律。设计:随机对照实验。地点和材料:实验地点为河北省医学科学院药理研究室。动物:健康雄性SD大鼠,体质量250~300g(由河北省实验动物中心提供)。方法:用线栓法建立大鼠大脑中动脉脑缺血(MCAO)模型后注射L-精氨酸和氨基胍,研究大鼠脑缺血后脑梗死体积变化;脑组织中一氧化氮、丙二醛含量和一氧化氮合酶、超氧化物歧化酶(SOD)活性的变化;缺血脑组织病理变化。主要观察指标:脑梗死体积;脑组织丙二醛含量;SOD活性;脑组织一氧化氮含量;NOS活性。结果:单独给予L-精氨酸犤(14.46±3.59)%犦或氨基胍犤(14.40±3.06)%犦均可明显缩小脑梗死体积犤假手术组为(22.10±3.98)%犦,明显降低缺血脑组织丙二醛含量,增强SOD活性;L-精氨酸还可明显增加缺血脑组织一氧化氮含量,氨基胍可明显抑制缺血脑组织NOS活性;L-精氨酸与氨基胍合用时,上述各项指标与缺血组比较无明显变化。结论:L-精氨酸与氨基胍分别单独应用,对脑缺血性损伤具有治疗作用,两药合用则无明显效果。
Background: It is always a hot research topic to find out the best medicine to treat cerebral ischemia, the optimal administration time and dosage with the dual action of nitric oxide. Objective: To observe the therapeutic effect of L-arginine and aminoguanidine on focal cerebral ischemic injury in rats and the role of nitric oxide in cerebral ischemic injury. Design: Randomized controlled experiment. Venue and Materials: The experimental site is the Pharmacology Laboratory of Hebei Academy of Medical Sciences. Animals: Healthy male SD rats, body weight 250 ~ 300g (provided by the Experimental Animal Center of Hebei Province). Methods: The rat middle cerebral artery occlusion (MCAO) model was established by thread occlusion. L-arginine and aminoguanidine were injected to study the changes of cerebral infarction volume after cerebral ischemia. The levels of nitric oxide, Aldehyde content and the activity of nitric oxide synthase and superoxide dismutase (SOD); pathological changes of ischemic brain tissue. MAIN OUTCOME MEASURES: Volume of cerebral infarction; MDA content in brain tissue; SOD activity; nitric oxide content in brain tissue; NOS activity. Results: L-arginine alone (14.40 ± 3.06)% 犦 or aminoguanidine 犦 (40 14.40 ± 3.06)% 单 significantly reduced the volume of cerebral infarction 犤 (22.10 ± 3.98)% 犤, which was significantly lower L-arginine can also significantly increase the content of nitric oxide in ischemic brain tissue, aminoguanidine can significantly inhibit the activity of NOS in ischemic brain tissue; L-arginine and amino Guanidine combined, the above indicators no significant change compared with the ischemic group.
