目的:探讨蝎毒多肽Ⅲ(PSV)和蛇毒多肽Ⅷ(KD)对Lewis肺癌(LLC)荷瘤小鼠肿瘤生长、肺部转移和一般状态的影响。方法:采用近交系C57BL/6小鼠,左腋下接种LLC细胞悬液建立肺转移模型,随机分为 5组,每天分别腹腔注射生理盐水(NS)、环磷酰胺(CTX)、高剂量PSV(PSV-H)、低剂量PSV(PSV-L)和KD,共 18 d。以原位瘤重、肺转移率和转移数为指标,检测不同浓度的PSV和KD对LLC生长和转移的抑制作用;以体重和脾系数为指标观察PSV和KD对带瘤小鼠一般状态的影响。结果:结果显示,PSV和KD对LLC原位肿瘤的抑制作用不明显(P>0.05),但它们对肿瘤肺转移却有非常明显的抑制作用PSV-H 0.33±0.2l,(P< 0.01);其免疫保护方面的作用也明显优于CTX。结论:蝎毒多肽Ⅲ和蛇毒多肽Ⅷ抑制原位瘤的作用缓慢,但是抑制肿瘤转移方面强于化疗药物CTX,并能够提高小鼠的生存质量。
Objective: To investigate the effects of PSV and KD on tumor growth, lung metastasis and general status in Lewis lung cancer (LLC) mice. Methods: The pulmonary metastasis model was established by inoculation of LLC cell suspensions in the left axilla of C57BL / 6 mice. The models were randomly divided into 5 groups: NS, CTX, PSV (PSV-H), low-dose PSV (PSV-L) and KD for 18 days. In situ tumor weight, lung metastasis rate and the number of metastasis as an indicator, the detection of different concentrations of PSV and KD on the growth and metastasis of LLC inhibition; weight and spleen coefficient as an indicator of PSV and KD on tumor-bearing mice general state influences. Results: The results showed that the inhibitory effect of PSV and KD on LLC in situ tumors was not significant (P> 0.05), but they had a significant inhibitory effect on tumor lung metastasis PSV-H 0.33 ± 0.2l, (P <0.01). Its immune protection was also significantly better than that of CTX. Conclusion: Scorpion venom peptide Ⅲ and snake venom peptide Ⅷ inhibited the effect of orthotopic tumor slowly, but the inhibition of tumor metastasis was stronger than that of chemotherapeutic drug CTX and could improve the quality of life of mice.
