本文报道应用Bond Elut C_(18)小柱或苯甲酸甲醇沉淀法提取茶碱作为样品的预处理,乙腈-水-乙酸或甲醇-水-乙酸为移动相,以β-羟乙基茶碱或苯甲酸作内标的两个反相等强度高效液相色谱测定法。方法学线性范围达277.5~550μmol/L(50~100μg/ml),回收率为87.3~103.3%,变异系数为1.7~6.7%,检测低限为2.78μmol/L和5.55μmol/L(0.5μg/ml和1.0μg/ml)。两法均具省时、简便和实用等特点,适于临床推广使用及药代动力学的研究。
In this paper, the extraction of theophylline using Bond Elut C_ (18) column or benzoic acid methanol precipitation as the sample pretreatment, acetonitrile-water-acetic acid or methanol-water-acetic acid as the mobile phase, β-hydroxyethyl theophylline or Benzoic acid as internal standard two anti-equal-intensity high-performance liquid chromatography. The linear range of the method was 277.5 ~ 550μmol / L (50 ~ 100μg / ml), the recovery was 87.3% ~ 103.3%, the coefficient of variation was 1.7 ~ 6.7% .The detection limit was 2.78μmol / L and 5.55μmol / L / ml and 1.0 μg / ml). Both methods are time-saving, simple and practical features, suitable for clinical use and pharmacokinetic studies.
