目的:检测Cyclin D1在腮腺多形性腺瘤及瘤旁涎腺组织中mRNA和蛋白水平的表达,探讨其在腮腺多形性腺瘤发生发展中的作用。方法:分别通过RT-PCR和Western blot方法检测Cyclin D1在原发腮腺多形性腺瘤与瘤旁涎腺组织中mRNA和蛋白水平的表达情况;另外采用免疫组化法检测Cyclin D1在原发腮腺多形性腺瘤以及瘤旁涎腺组织石蜡组织标本中蛋白水平的表达情况。结果:RT-PCR和Western blot检测结果显示,CyclinD1在组织标本中mRNA和蛋白水平不仅在于肿瘤组织中有较强的表达,而且瘤旁涎腺组织中Cyclin D1也有一定量的表达。免疫组化检测结果显示,Cyclin D1在腮腺多形性腺瘤组织中有较高的表达,而在瘤旁涎腺组织中未见明显表达。结论:Cyclin D1可能参与腮腺多形性腺瘤的发生发展,有助于为临床诊断治疗提供一定的参考依据。
OBJECTIVE: To detect the expression of Cyclin D1 mRNA and protein in parotid pleomorphic adenoma and para-tumor salivary glands, and to explore the role of Cyclin D1 in the development of parotid pleomorphic adenoma. Methods: The mRNA and protein expressions of Cyclin D1 in primary parotid gland adenoma and adjacent para-salivary gland tissues were detected by RT-PCR and Western blot respectively. In addition, the expression of Cyclin D1 in the primary parotid gland was detected by immunohistochemistry Pleomorphic adenoma and para-tumor salivary gland paraffin tissue specimens of protein expression. Results: The results of RT-PCR and Western blot showed that the mRNA and protein levels of CyclinD1 in the tissue samples not only showed strong expression in tumor tissues, but also showed a certain amount of expression of Cyclin D1 in para-tumor salivary glands. The results of immunohistochemistry showed that Cyclin D1 was highly expressed in the pleomorphic adenoma tissues, but not in the paraneoplastic salivary gland tissues. Conclusion: Cyclin D1 may be involved in the development of parotid pleomorphic adenoma, which will be helpful to provide some reference for clinical diagnosis and treatment.
