目的:观察丹参酮ⅡA对兔动脉粥样硬化斑块胶原含量的影响,并对其机制进行初步探讨。方法:日本大耳兔随机分为对照组和丹参酮ⅡA(7、20、60mg/kg)4组,高胆固醇饮食10周建立兔动脉粥样硬化(atherosc lerosis,AS)病变模型后药物干预4周,取主动脉内膜组织分别采用W oessner方法、免疫组织化学方法和realtim e RT-PCR法检测斑块组织中胶原含量,MMP-1蛋白,IL-10、IL-1 mRNA表达,W estern-b lot方法检测斑块组织中PPAR-γ蛋白表达。结果:丹参酮ⅡA低剂量能明显提高斑块中胶原蛋白含量、PPAR-γ蛋白和IL-10 mRNA表达,明显降低斑块组织中MMP-1和IL-1 mRNA的表达;中、高剂量组能显著提高斑块中胶原蛋白含量、PPAR-γ蛋白和IL-10 mRNA表达,显著降低斑块组织中MMP-1和IL-1 mRNA的表达。结论:丹参酮ⅡA能提高动脉粥样硬化斑块中胶原蛋白含量,其机制可能与抑制炎症反应,降低MMP-1表达有关。
Objective: To observe the effect of tanshinone Ⅱ A on the collagen content of atherosclerotic plaque in rabbits, and to explore its mechanism. Methods: Japanese white rabbits were randomly divided into 4 groups: control group and tanshinone ⅡA (7, 20 and 60 mg / kg) groups. Rabbits with atherosclerotic atherosclerosis (AS) model were established by hypercholesterolemia for 10 weeks. The aortic intima tissues were taken out to detect the collagen content, the expression of MMP-1, IL-10 and IL-1 mRNA in the plaque tissue by W oessner method, immunohistochemistry and realtim e RT- b lot method to detect the expression of PPAR-γ protein in plaque tissue. Results: Low doses of tanshinone ⅡA could significantly increase the plaque collagen content, PPAR-γ protein and IL-10 mRNA expression, significantly reduce the expression of MMP-1 and IL-1 mRNA in plaque tissue; Significantly increased plaque collagen content, PPAR-γ protein and IL-10 mRNA expression, significantly reduced plaque tissue MMP-1 and IL-1 mRNA expression. Conclusion: Tanshinone ⅡA can increase the content of collagen in atherosclerotic plaque. Its mechanism may be related to inhibiting the inflammatory reaction and decreasing the expression of MMP-1.
