目的研究子宫内膜异位症对Wistar大鼠胚胎种植期子宫内膜整合素α_vβ_3、血管内皮生长因子(VEGF)基因的表达及雌、孕激素水平与不孕的关系。方法采用同种异体移植方法建立Wistar大鼠子宫直肠陷凹、腹壁外子宫内膜异位症动物模型36只,空白对照20只,建模成功后投放雄鼠,受孕4.5 d后麻醉采血,取模型大鼠在位子宫内膜组织,进行透射电镜观察,ELISA检测血清中雌激素、孕激素的含量,同时运用荧光定量PCR技术,定量分析整合素α_vβ_3以及VEGF基因在其子宫内膜中的表达。结果早孕各组大鼠子宫内膜α_vβ_3、VEGF基因的表达水平均高于模型组和空白未孕组;早孕能提高模型组α_vβ_3、VEGF基因的表达但均低于空白受孕组(P<0.05),且子宫直肠凹陷子宫内膜异位症模型大鼠的表达水平较腹壁外子宫内膜异位症模型大鼠低(P<0.05);子宫直肠凹陷子宫内膜异位症模型大鼠血清雌激素水平低于空白受孕组(P<0.05),但孕激素较空白受孕组高(P<0.05)。结论不同程度的子宫内膜异位症均能降低Wistar大鼠窗口期整合素α_vβ_3、VEGF基因的表达,使其达不到正常胚胎种植期的水平是导致不孕的重要原因。
Objective To study the relationship between endometriosis endometrial integrin α_vβ_3 and vascular endothelial growth factor (VEGF) gene expression and estrogen and progesterone levels in infertile women during Wistar rat embryo implantation. Methods Thirty-six cases of Wistar rat uterine rectum and extra-abdominal wall endometriosis were established by allograft method. Twenty rabbits were randomly divided into control group and control group. The model rats were placed in endometrial tissue and examined by transmission electron microscopy. The levels of estrogen and progesterone in serum were detected by ELISA. The expression of integrin α_vβ_3 and VEGF gene in endometrium was quantitatively analyzed by fluorescence quantitative PCR . Results The expression levels of α_vβ_3 and VEGF in endometrium in early pregnancy were higher than those in model group and blank non-pregnant group. The expression of α_vβ_3 and VEGF in model group were significantly higher than those in blank control group (P <0.05) , And the expression level of uterine rectum in endometriosis model rats was lower than that in abdominal wall endometriosis model rats (P <0.05). The serum estradiol level in uterine rectal depression and endometriosis model rats The level of hormone was lower than that of the blank control group (P <0.05), but the level of progesterone was higher than that of the blank control group (P <0.05). Conclusion Different degrees of endometriosis can reduce the expression of integrin α_vβ_3 and VEGF in the window of Wistar rats, which can not reach the level of normal embryo implantation, which is the important reason of infertility.
