目的建立一种快速、灵敏的方法间接测定人血浆和尿样中的羟乙基淀粉浓度。方法血浆和尿样经三氯乙酸沉淀蛋白,丙酮沉淀羟乙基淀粉后,以三氟乙酸水解羟乙基淀粉,采用己糖激酶法测定水解产物单糖,从而计算血浆和尿中羟乙基淀粉的浓度。结果测定血浆中羟乙基淀粉方法的线性范围为0.25~10.0 mg.mL-1,其涵盖实际剂量下血药浓度范围。方法确证各结果均符合相关标准。实验每天可以分析150个样品。结论本分析方法测试时间短,灵敏度高,血浆用量少,适用于人血浆和尿样中羟乙基淀粉的测定及其药动学研究。
Objective To establish a rapid and sensitive method for the indirect determination of hydroxyethyl starch in human plasma and urine samples. Methods After plasma and urine samples were precipitated with trichloroacetic acid and hydroxyethyl starch was precipitated by acetone, hydroxyethyl starch was hydrolyzed by trifluoroacetic acid and the hydrolyzate monosaccharide was determined by hexokinase method, and then the content of hydroxyethyl The concentration of starch. Results The linear range of the method for determination of hydroxyethyl starch in plasma was 0.25-10.0 mg.mL-1, which covered the range of plasma concentrations at the actual dose. Methods confirm that the results are in line with the relevant standards. The experiment can analyze 150 samples daily. Conclusion The method has short test time, high sensitivity and low plasma dosage. It is suitable for the determination of hydroxyethyl starch in human plasma and urine samples and its pharmacokinetic study.
