目的探讨不同剂量下,黄连及其复方黄连解毒汤的抗炎效果及抑菌作用,为临床合理使用黄连的剂量提供初步的实验依据。方法采用二甲苯致小鼠耳肿胀模型、醋酸致小鼠腹腔毛细血管通透性增加模型及鲍氏志贺氏菌模型,观察体内抗炎及体外抑菌作用。黄连(单)1~4组及黄连解毒汤(复)1~4组分别给予含黄连生药3.0,1.5,0.75,0.3g·kg-1水煎药液;阳性药组给予阿司匹林0.2g·kg-1;正常组和模型组给予等体积蒸馏水。均灌胃给药,连续5d,每日1次。分别于末次给药后30,40min用二甲苯涂抹小鼠左耳两面、醋酸腹腔注射造模,称量两耳片质量、检测腹腔涮洗液吸光度值。将不同浓度的黄连及黄连解毒汤水煎液(1~5组)制成药敏纸片,贴于涂布鲍氏志贺氏菌的mH琼脂平板上,24h后观察抑菌环大小。结果模型组小鼠耳肿胀度明显增加(P<0.01),与模型组比较,阳性药组、单1~4组、复1~3组,可明显抑制二甲苯所致小鼠耳肿胀度(P<0.01,P<0.05);除单1与单4外,单味药各组间无明显差异,复方各组间无明显差异。模型组小鼠腹腔涮洗液吸光度值(A)明显增加(P<0.01),与模型组比较,阳性药组、单1~3组、复1~3组,可明显降低腹腔涮洗液A值,抑制醋酸致腹腔毛细血管通透性增加(P<0.01,P<0.05);单1~3组组间、复1~3组组间均无明显差异。单味及复方各剂量组对鲍氏志贺氏菌均有抑制作用,抑菌环随剂量增大而增大;单3~5、复3~5抑菌强度无差别。结论黄连及其复方黄连解毒汤均具有抗炎、抗菌作用,在临床常用剂量范围内,其作用效果与剂量无明显正相关性。
Objective To investigate the anti-inflammatory and antibacterial effects of Coptis chinensis and its compound Huanglianjiedu Decoction under different doses, and to provide a preliminary experimental basis for the clinical rational use of Coptis chinensis. Methods Xylene-induced mouse ear swelling model, acetic acid-induced mouse models of increased peritoneal capillary permeability and Shigella boyneder model were observed in vivo anti-inflammatory and in vitro antibacterial activity. Coptis chinensis (single) 1 ~ 4 group and Huanglian Jiedu Decoction (complex) 1 ~ 4 group were treated with Coptis crude drug 3.0,1.5,0.75,0.3g · kg-1 decoction; positive drug group given aspirin 0.2g · kg -1; normal group and model group were given equal volume of distilled water. All gavage administration, continuous 5d, 1 day. At the end of 30 and 40min after the last administration, both sides of the left ear of mice were smeared with xylene. The rats were injected intraperitoneally with acetic acid and the quality of the two ears was weighed. The absorbance of abdominal washings was measured. Different concentrations of Coptis and Coptis decoction decoction (1 to 5 groups) made of drug-sensitive paper, affixed to Shigella flexneri mH agar plate, 24h inhibition inhibition ring size. Results Compared with the model group, the positive drug group, single group 1 ~ 4, and complex 1 ~ 3 groups significantly reduced the ear edema in mice induced by xylene (P <0.01) P <0.01, P <0.05). There was no significant difference between single groups and single groups except for single 1 and single 4, but there was no significant difference between each group. Compared with the model group, the positive group, single group 1 ~ 3, complex 1 ~ 3 group, can significantly reduce abdominal atrophy rinse solution A (P <0.01) (P <0.01, P <0.05). There was no significant difference between groups 1 to 3 and between groups 1 and 3 when acetic acid was used to inhibit peritoneal capillary permeability. Monosodium glutamate group and compound dose group had inhibitory effect on Shigella blastulis, and antibacterial ring increased with dosage increasing. There was no difference in the antibacterial activity of single 3 ~ 5 and complex 3 ~ 5. Conclusions Coptis and its compound Huanglian Jiedu Decoction have anti-inflammatory and antibacterial effects. There is no significant positive correlation between the effect and dose of Coptis chinensis and its compound Huanglian Jiedu Decoction in the range of common clinical dose.
