目的建立短暂全脑缺血的成年大鼠视网膜微循环缺血模型。方法选取实验动物为Wistar雄性大鼠5只,采用Pulsinelli经典的大鼠四血管结扎模型,烧灼双侧椎动脉后,结扎双侧颈总动脉,缺血10 min后解除夹闭的缺血装置,恢复脑血流。对成活大鼠模型制成后,经麻醉、散瞳,行眼底荧光造影和病理形态检查,观察成年大鼠短暂全脑缺血的视网膜改变。结果全部5只短暂全脑缺血实验大鼠均显示视网膜微循环缺血,缺血大鼠眼底荧光造影:视网膜血管放射状充盈不完全;病理形态改变:视网膜血管不同程度充血,视网膜节细胞层少数细胞胞浆空泡变性。结论构建了视网膜微循环缺血实验模型,为今后临床药物的相关研究提供了理想的实验动物模型。
Objective To establish a rat model of retinal microcirculation ischemia with transient global cerebral ischemia. Methods Five experimental Wistar male rats were selected. Pulsinelli model was used to establish the model of four-vessel occlusion in rats. Bilateral vertebral arteries were cauterized and bilateral common carotid arteries were ligated. Ischemic devices were released after 10 min of ischemia. Restore cerebral blood flow. After the model of the living rat was made, the retinal changes of transient global cerebral ischemia in adult rats were observed under anesthesia, mydriasis, fundus fluorescein angiography and pathological examination. Results All 5 rats with transient global cerebral ischemia showed retinal microcirculation ischemia and fundus fluorescein angiography in rats: Radial filling of retinal vessels was not complete; Pathological changes: retinal blood vessels were hyperaemic to varying degrees, few retinal ganglion cell layers Cytoplasmic vacuolar degeneration. Conclusion The experimental model of retinal microcirculation ischemia is established, which provides an ideal experimental animal model for the related research of clinical drugs in the future.
