目的:观察血小板源性生长因子A链(PDGF-A)、胰岛素样生长因子-Ⅰ(IGF-Ⅰ)在动脉粥样硬化斑块中的表达及分布差异,探讨两者在动脉粥样硬化发展过程中的作用。方法:应用免疫组织化学技术测定31人股动脉粥样斑块中同一部位PDGF-A、IGF-Ⅰ的表达。结果:闭塞性动脉硬化症患者股动脉粥样斑块中两种生长因子均呈强阳性表达,而正常人动脉壁中两者仅在中膜有微量表达,两者比较有显著差异,但两种因子之间比较无明显差异。在分布特点上,两种生长因子主要分布在平滑肌细胞密集的部位,即新生斑块区,但又有不同,PDGF-A在泡沫细胞区域也有分布,IGF-Ⅰ在泡沫细胞区域仅有少量分布。结论:PDGF-A和IGF-1在动脉粥样斑块中的表达增高与AS及ASO的发生密切相关;其分布区域的差异与其表达细胞的种类不同有关,但都主要由增殖的平滑肌细胞表达。
Objective: To observe the expression and distribution of platelet-derived growth factor A chain (PDGF-A) and insulin-like growth factor-Ⅰ (IGF-Ⅰ) in atherosclerotic plaque and to explore their roles in the development of atherosclerosis The role of the process. Methods: Immunohistochemistry was used to detect the expression of PDGF-A and IGF-Ⅰ in the same site of 31 human atherosclerotic plaques. Results: Both of the two growth factors in femoral atherosclerotic plaques showed a strong positive expression in patients with occlusive arteriosclerosis, whereas both normal and normal human arteries showed only slight expression in the mesothelial lining, but there was a significant difference between the two No significant difference between the factors. In distribution characteristics, the two growth factors are mainly distributed in the smooth muscle cell dense parts, namely the new plaque area, but there are differences, PDGF-A is also distributed in the foam cell area, IGF-I in the foam cell area only a small amount of distribution . CONCLUSION: The increased expression of PDGF-A and IGF-1 in atherosclerotic plaque is closely related to the development of AS and ASO. The distribution of PDGF-A and IGF-1 is related to the different types of their expressed cells, but they are mainly expressed by proliferating smooth muscle cells .
