目的 :探讨甘氨酸和多粘菌素B在体内对内毒素诱导的家兔急性期反应的影响。方法 :复制内毒素诱导急性期反应模型 ,其中实验组预先给予半剂量的甘氨酸 /多粘菌素B合剂 ,当家兔的体温上升至高峰后 1h采血 ,进行白细胞计数、血清C -反应蛋白、微量元素 (Cu2 + 、Fe2 + 、Zn2 + )的测定。结果 :预先给予半剂量的甘氨酸 /多粘菌素B合剂可以显著抑制内毒素诱导急性期反应 ,包括对家兔体温、C -反应蛋白以及微量元素 (Cu2 + 、Fe2 + 、Zn2 + )的变化的抑制等 (P <0 0 5 ) ,并且其效果与预先给予全剂量多粘菌素B无显著性差异 (P >0 0 5 )。结论 :在内毒素性疾病治疗中 ,选择分别作用于内毒素不同结构部位的两种拮抗剂组成合剂 ,可以利用某些拮抗剂的不同特点相互取长补短 ,从多个环节上拮抗内毒素的生物学作用 ,降低单一药物的剂量 ,减轻毒副反应。
Objective: To investigate the effect of glycine and polymyxin B on endotoxin-induced acute phase reaction in rabbits in vivo. Methods: The acute phase response model was induced by endotoxin. The experimental group was given a half dose of glycine / polymyxin B mixture. When rabbits’ body temperature rose to the peak, blood samples were taken at 1 hour. The white blood cell count, serum C - reactive protein, Determination of trace elements (Cu2 +, Fe2 +, Zn2 +). Results: Pretreatment with glycine / polymyxin B at half dose significantly inhibited the acute phase reaction induced by endotoxin, including the change of body temperature, C - reactive protein and trace elements (Cu2 +, Fe2 +, Zn2 +) in rabbits (P <0 05), and its effect was not significantly different from that of the pretreatment with full dose of polymyxin B (P> 0.05). Conclusion: In the treatment of endotoxin disease, we choose two kinds of antagonistic agents acting on different structural parts of endotoxin respectively, which can take advantage of the different characteristics of certain antagonists to complement each other and antagonize the biology of endotoxin from multiple aspects The role of reducing the dose of a single drug to reduce side effects.
