目的:观察补肾方对Con A诱导的慢性肝损伤小鼠肝组织Th1/Th2的影响并探讨其作用机制。方法:将150只Balb/c小鼠,根据完全随机原则分为5组,即分别为正常组,模型组,补肾方组,秋水仙碱组,甘草酸苷组,每组30只。Con A用无菌0.9%氯化钠溶液配制成1.25mg/m L浓度,小鼠按0.1m L/kg标准尾静脉注射给药,正常组给予等量的0.9%氯化钠溶液尾静脉注射。每周1次,连续6周,构建Con A诱导慢性肝损伤小鼠模型。造模同时分别给予补肾方、秋水仙碱、复方甘草酸苷胶囊(凯因甘乐)灌胃。观察各组小鼠肝功能,HE染色,流式细胞术检测小鼠肝脏内Th1、Th2细胞数量。结果:与正常组比较,模型组血清ALT、AST明显升高(P<0.05),补肾方组、甘草酸苷组血清ALT、AST较模型组降低(P<0.05)。流式细胞检测发现补肾方组、秋水仙碱组、Th1细胞数量都明显少于模型组(P<0.05);补肾方组、秋水仙碱组和甘草酸苷组Th2细胞数量都明显少于模型组(P<0.05),Th1/Th2比值多于模型组(P<0.05)。结论:补肾方能够降低Con A诱导的小鼠慢性肝损伤模型中肝脏内浸润的Th1、Th2细胞数目,提高Th1/Th2的比值,降低血清学指标,减轻肝脏病理改变,减少肝组织炎性细胞的浸润。
Objective: To observe the effect of Bushen Recipe on the Th1 / Th2 in liver tissue of mice with chronic liver injury induced by Con A and explore its mechanism. Methods: 150 Balb / c mice were divided into 5 groups according to the principle of complete randomness, namely, normal group, model group, Bushen Fang group, colchicine group and glycyrrhizin group, 30 in each group. Con A was prepared to a concentration of 1.25 mg / mL with a sterile 0.9% sodium chloride solution. The mice were injected with 0.1 m L / kg of standard tail vein and the normal group with 0.9% sodium chloride solution by tail vein injection . Once a week for 6 weeks, Con A-induced chronic liver injury in mice model. At the same time modeling were given kidney side, colchicine, compound glycyrrhizin capsules (Caine Gan Le) gavage. The liver function of each group was observed, HE staining and flow cytometry were used to detect the number of Th1 and Th2 cells in the liver of mice. Results: Compared with the normal group, serum ALT and AST in model group were significantly increased (P <0.05). The serum ALT and AST in Bushenfang group and glycyrrhizin group were lower than those in model group (P <0.05). Flow cytometry showed that the number of Th2 cells in Bushenfang group, colchicine group and Th1cells was significantly less than that in model group (P <0.05). The numbers of Th2cells in Bushenfang group, colchicine group and glycyrrhizin group were significantly less than those in model group Group (P <0.05), Th1 / Th2 ratio more than the model group (P <0.05). Conclusion: Bushen Recipe can reduce the number of Th1 and Th2 cells infiltrating into the liver induced by Con A in mice model of chronic liver injury, increase the ratio of Th1 / Th2, decrease the serological parameters, alleviate the pathological changes of the liver and reduce the number of inflammatory cells Infiltration.
