本研究采用体外组织培养法观察α-促黑素细胞激素(α-MSH)对白细胞介素-1_β/(IL-1_β)刺激家兔下丘脑组织释放环—磷酸腺苷(cAMP)的影响。结果显示,IL-1_β与下丘脑组织培养40min后,培养上清液中cAMP含量显著高于未加IL-1_β的对照组(P<0.01);而预先(前20min)加入α-MSH再加入同量IL-1_β培养40min,则培养上清液的cAMP含量接近对照组水平(P>0.05)。我们认为,α-MSH抑制下丘脑cAMP的产生,可能是其解热的重要机制之一。
In this study, the effect of α-MSH on interleukin-1β (IL-1β) -induced cAMP release in hypothalamus of rabbits was observed by in vitro tissue culture. The results showed that the content of cAMP in supernatant of IL-1β and hypothalamus was significantly higher than that of the control group without IL-1β (P <0.01) after pre-incubation (before 20min) The same amount of IL-1_β cultured 40min, the culture supernatant cAMP content close to the control group (P> 0.05). We believe that α-MSH inhibition of hypothalamic cAMP production may be one of its important mechanisms of antipyretic.
