探讨重组人粒－巨噬细胞集落刺激因子的分离、纯化工艺。方法；经破菌溶解包含体、复性等粗提纯后，以三步色谱法精制纯化，对终产品进行纯度、生物活性、热原及急性毒性检测。结果：终产品为单一条带、单一峰，生物活性为１．５９ ×１０~７～１．８６×１０~７ＩＵ／ｍｇ，热原测定及急性毒性试验符合要求。结论：此工艺纯化的产品纯度高、生物活性好，有一定的应用价值。
To investigate the isolation and purification of recombinant human granulocyte-macrophage colony-stimulating factor. Methods: After the crude lysates were dissolved in inclusion bodies and refolded, the purified product was purified by three-step chromatography. The purity, bioactivity, pyrogen and acute toxicity of the final product were tested. Results: The final product was a single band with a single peak. The bioactivity was 1.59 × 10 ~ 7 ~ 1.86 × 10 ~ 7IU / mg. The pyrogen determination and acute toxicity test met the requirements. Conclusion: The purified product of this process has high purity, good biological activity and has certain application value.
