为探讨压力负荷增高性心肌肥厚大鼠心肌胶原网络的重构及其与左室功能的关系。采用腹主动脉结扎术致大鼠心肌肥厚;心脏离体灌注测定左室功能;VG染色和图像处理观察心肌胶原网络重构。结果显示手术组大鼠术后2周即出现明显左室肥厚;与假手术组比较,左室心肌总胶原容积百分比(CVF-T)于术后2周即呈增高趋势(P<0.05),术后4,8周CVF-T和无小血管视野CVF(CVF-NV)均显著高于假手术组;手术组大鼠术后4周左室功能出现明显减退,CVF-NV,CVF-T与左室功能指标,尤其舒张功能指标呈明显负相关。结论:压力负荷增高性心肌肥厚时伴有心肌胶原网络的重构,心肌胶原网络重构与心功能障碍,尤其是舒张功能障碍有明显关系。
To investigate the remodeling of myocardial collagen network in hypertensive cardiac hypertrophy and its relationship with left ventricular function. Cardiac hypertrophy was induced by abdominal aortic ligation in rats. Left ventricular function was measured by isolated heart perfusion. VG staining and image processing were used to observe myocardial collagen network remodeling. The results showed that left ventricular hypertrophy was found in the operation group 2 weeks after operation. Compared with the sham operation group, the CVF-T of left ventricular myocardium increased at 2 weeks after operation (P <0.05 ), CVF-T and CVF-NV at 4 and 8 weeks after operation were significantly higher than those in sham-operated group. The left ventricular function was significantly decreased 4 weeks after operation in rats in operation group. CVF-NV and CVF -T and left ventricular function index, especially diastolic function index was significantly negatively correlated. CONCLUSION: Myocardial collagen network remodeling is associated with myocardial hypertrophy in stress load. The remodeling of myocardial collagen network is obviously associated with cardiac dysfunction, especially diastolic dysfunction.
