目的:研究黄连解毒汤对糖尿病大鼠肝脏核转录因子-κB(NF-κB),肿瘤坏死因子-α(TNF-α)表达的影响,探讨黄连解毒汤治疗糖尿病的机制。方法:实验采用高脂饮食加小剂量链脲佐菌素(STZ)复制大鼠糖尿病模型,连续给黄连解毒汤(11.5,5.7,2.85 g.kg-1)8周后测定空腹血糖(FGB)、总胆固醇(CHO)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDLC)、高密度脂蛋白胆固醇(HDLC)含量,同时计算肝脏和胰腺质量指数;采用免疫组织化学的方法检测大鼠肝脏中NF-κB,TNF-α含量。结果:黄连解毒汤高剂量组能显著降低大鼠血清中CHO,TG,LDLC含量,各剂量组均升高HDLC含量,明显降低胰腺的质量指数;大鼠肝脏NF-κB,TNF-α的表达明显低于模型组。结论:黄连解毒汤对高脂饮食加小剂量STZ所致糖尿病大鼠有一定的治疗作用,其机制可能是通过调节NF-κB,TNF-α蛋白的表达,从而抑制了炎症信号的通路。
Objective: To investigate the effect of Huanglian Jiedu Decoction on the expression of NF-κB and TNF-α in the liver of diabetic rats, and to explore the mechanism of Huanglian Jiedu Decoction in the treatment of diabetes mellitus. Methods: Experimental diabetic rats were given a high-fat diet plus a small dose of streptozotocin (STZ), and the fasting blood glucose (FGB), fasting blood glucose (FBG) , Total cholesterol (CHO), triglyceride (TG), low density lipoprotein cholesterol (LDLC) and high density lipoprotein cholesterol (HDLC) were measured. The liver and pancreas mass index were calculated. Immunohistochemistry Liver NF-κB, TNF-α content. Results: The Huanglian Jiedu Decoction high-dose group could significantly reduce the content of CHO, TG and LDLC in rats serum, the content of HDLC in each dose group was increased, the mass index of pancreas was significantly decreased; the expression of NF-κB and TNF-α Obviously lower than the model group. CONCLUSION: Huanglian Jiedu Decoction has some therapeutic effects on rats fed with high-fat diet and low-dose STZ, and its mechanism may be through inhibiting the expression of NF-κB and TNF-α, thereby inhibiting the pathway of inflammatory signals.
