目的:对不同代次的脂肪干细胞表面抗原进行流式细胞术分析,为组织工程学的种子细胞的选用提供理论依据。方法选择第5代,第6代,第8代,第10代脂肪干细胞分别消化收集,封闭,并用CD73,CD90,CD105荧光染色,冰上避光孵育,离心洗涤后过滤,冰上备用,流式细胞仪检测。结果 CD73,CD90,CD105三种荧光抗体标记皆为第5代的阳性率最高,第10代阳性率最低。结论表明脂肪干细胞的干细胞特征会随着传代次数的增加而减少,传代次数少更有利于组织工程学的研究。“,”Objective Analysis of positive express of dif erent passages Human Adipose-derived Stem Cel s by Flow Cytometry. Methods The cel s biological characteristics of passage 5, passage 6, passage 8 and passage 10 which grew wel were digested and marked with CD73,CD90,CD105. Positive expression of CD73, CD90,CD105 were detected by Flow cytometry. Results The highest positive expression of CD73,CD90,CD105was the Human Adipose-derived Stem Cel s in passaged 5, the least positive expression of three antibody was the ADSCs in passaged 10. Conclusion The fewer the times of passages the bet er for the study of skin tissue engineering.
