目的:研究渐尖毛蕨总黄酮对慢性镉暴露诱导大鼠肾脏损伤的影响。方法:采用暴露于含250 mg/L氯化镉饮水16 w的方式诱导大鼠慢性肾损伤模型,灌胃渐尖毛蕨总黄酮干预。实验周期结束后检测大鼠血肌酐(Cr)和尿素氮(BUN)含量、肾脏胶原沉积情况及血清超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)活性和丙二醛(MDA)水平,并检测肾脏TGF-β1和微小RNA(miR)-29a/b/c的表达。结果:渐尖毛蕨总黄酮能显著改善镉诱导的大鼠肾脏胶原沉积,降低血清Cr、BUN、MDA水平,提高血清SOD和GSH-Px活性,抑制肾组织TGF-β1表达并上调miR-29a/b/c表达。结论:渐尖毛蕨总黄酮对慢性镉暴露诱导的大鼠肾脏损伤具有潜在的抑制作用,其作用机制与抑制氧化应激,调控肾脏TGF-β1及miR-29a/b/c表达有关。
OBJECTIVE: To study the effects of total flavonoids of Pseudostellariae present on the kidney injury induced by chronic exposure to cadmium in rats. Methods: Chronic renal injury model was induced in rats exposed to 250 mg / L cadmium chloride drinking water for 16 weeks, and the total flavonoids were isolated from ginseng. Serum creatinine (Cr) and blood urea nitrogen (BUN), renal collagen deposition and the activities of serum SOD and GSH-Px in the rats were measured after the end of the experimental period Malondialdehyde (MDA) levels were measured and the expressions of TGF-β1 and miR-29a / b / c in kidney were detected. Results: The total flavonoids of Rhododendron fortunei could significantly improve cadmium-induced collagen deposition in rat kidneys, decrease serum levels of Cr, BUN and MDA, increase serum SOD and GSH-Px activities, inhibit TGF-β1 expression and up-regulate miR-29a / b / c expression. CONCLUSION: The total flavonoids of Rhododendron mongolicus could potentially inhibit the kidney injury induced by chronic exposure to cadmium. The mechanism of action is related to inhibiting oxidative stress and regulating the expression of TGF-β1 and miR-29a / b / c in kidney.
