目的:探讨Survivin在卵巢癌顺铂耐药中的作用。方法:应用RT-PCR和Western Blot检测卵巢癌顺铂敏感细胞株SKOV3和耐药株SKOV3/DDP中survivin表达,MTT法检测细胞增殖抑制率,流式细胞技术检测卵巢癌细胞的凋亡率,Survivin反义寡核苷酸被脂质体转染至SKOV3/DDP细胞中。结果:SKOV3/DDP中Survivin mRNA的相对表达强度为(1.21±0.59),显著高于SKOV3细胞中(0.54±0.23),差异有统计学意义(P<0.05)。Survivin蛋白在两种细胞的相对表达强度分别是SKOV3细胞(0.87±0.48)和SKOV3/DDP细胞(2.07±1.01),差异有统计学意义(P<0.05)。Survivin-ASOD可降调卵巢癌细胞中Survivin的表达,并显著增加细胞对顺铂敏感性。结论:卵巢癌细胞对顺铂耐药可能与SKOV3/DDP细胞中Survivin表达增加有关。
Objective: To investigate the role of Survivin in cisplatin-resistant ovarian cancer. Methods: Survivin expression was detected by RT-PCR and Western Blot in cisplatin-sensitive ovarian cancer cell line SKOV3 and drug-resistant cell line SKOV3 / DDP. MTT assay was used to detect the cell proliferation inhibition rate. Flow cytometry was used to detect the apoptosis rate of ovarian cancer cells. Survivin antisense oligonucleotides were liposome transfected into SKOV3 / DDP cells. Results: The relative expression intensity of Survivin mRNA in SKOV3 / DDP cells was (1.21 ± 0.59), which was significantly higher than that in SKOV3 cells (0.54 ± 0.23), the difference was statistically significant (P <0.05). The relative expression intensity of Survivin protein in SKOV3 cells (0.87 ± 0.48) and SKOV3 / DDP cells (2.07 ± 1.01) was statistically significant (P <0.05). Survivin-ASOD can down-regulate the expression of Survivin in ovarian cancer cells and significantly increase the sensitivity of cells to cisplatin. Conclusion: The resistance of ovarian cancer cells to cisplatin may be related to the increased expression of Survivin in SKOV3 / DDP cells.
