目的探讨Survivin、细胞周期素依赖激酶4(Cyclin-dependent kinase 4,CDK4)、细胞增殖相关抗原(Ki-67)在儿童急性白血病(AL)中的表达及其相关性,同时观察其与AL临床分型和预后的关系。方法应用免疫组织化学链霉素亲和素-生物素-过氧化酶复合物(SABC)方法检测37例AL患儿骨髓细胞Survivin、CDK4、Ki-67表达,对照组10例为非恶性血液病且骨髓正常的儿童。结果37例AL患儿中,Survivin表达阳性率为45.9%,CDK4表达阳性率为56.8%,Ki-67表达阳性率为40.5%,均显著高于正常骨髓组织(P均<0.05);Survivin与CDK4表达呈强相关(P=0.007),Survivin与Ki-67表达呈强相关(P=0.008)。结论Survivin、CDK4、Ki-67在AL患儿中均过度表达,共同参与AL患儿白血病细胞凋亡和增殖的调控过程。联合检测Survivin、CDK4、Ki-67有助于AL的诊断、预后及疗效判断。
Objective To investigate the expression and correlation of Survivin, CDK4 and Ki-67 in children with acute leukemia (AL) and to observe its relationship with AL clinical Relationship between typing and prognosis. Methods Immunohistochemical streptavidin-biotin-peroxidase complex (SABC) method was used to detect the expression of Survivin, CDK4 and Ki-67 in bone marrow cells of 37 children with AL. The control group of 10 patients with non-hematological malignancies And normal bone marrow children. Results The positive rate of Survivin was 45.9%, the positive rate of CDK4 was 56.8%, the positive rate of Ki-67 was 40.5% in 37 children with AL, which were significantly higher than those in normal bone marrow (all P <0.05) CDK4 expression was strongly correlated (P = 0.007), Survivin and Ki-67 expression was strongly correlated (P = 0.008). Conclusions Survivin, CDK4 and Ki-67 are overexpressed in children with AL and participate in the regulation of apoptosis and proliferation of leukemia cells in children with AL. Combined detection of Survivin, CDK4, Ki-67 contributes to the diagnosis, prognosis and curative effect of AL.
