通过静电纺丝技术制备聚苯乙烯马来酸酐超细纳米纤维,并采用冰冻切片法将长纤维切割为长度分别为20、50、80μm的短纤维。研究了HepG2肝癌细胞在不同长度短纤维支架上的生长、形貌、白蛋白分泌和尿素的合成、代谢相关酶的表达。结果表明,HepG2细胞在短纤维上可保持增殖、良好成球,并能维持较好的功能,且在50μm的短纤维上形貌和功能最佳。这就建立了一种良好的体外三维肿瘤模型,并能更加精确地模拟体内肿瘤真实情况,为后续的抗癌药物测试做好准备。
Poly (styrene maleic anhydride) ultrafine nanofibers were prepared by electrospinning technique, and the long fibers were cut into 20, 50 and 80μm staple fibers by cryo-sectioning. The growth, morphology, albumin secretion and urea synthesis of HepG2 hepatocarcinoma cells on different lengths of short fiber scaffolds were studied, and the expression of metabolic enzymes was studied. The results showed that HepG2 cells could maintain proliferation on the short fibers, spheroidize well and maintain good functions. The morphology and function of HepG2 cells on the 50μm short fibers were the best. This establishes a good three-dimensional tumor model in vitro and can more accurately mimic the true condition of the tumor in vivo for future anticancer drug testing.
