目的 探讨实验性自身免疫性脑脊髓炎 (EAE)大鼠脑血管内皮细胞表面分子的改变。 方法 用免疫组织化学方法检测EAE大鼠脑血管内皮细胞上血管细胞粘附分子 1(VCAM 1)和主要组织相容性复合体 (MHC)Ⅱ类分子的表达。用MTT(四甲基偶氮唑盐 )法检测不同发病级别大鼠血清TNF和IFN γ的水平。 结果 EAE大鼠脑血管内皮细胞较对照有诱导表达的VCAM 1和MHCⅡ类分子 ,血清中存在TNF和IFN γ活性。 结论 中枢神经系统炎症状态下 ,脑血管内皮细胞表面分子发生改变 ,提示它们在EAE发病中可能起重要作用
Objective To investigate the molecular changes of cerebrovascular endothelial cells in experimental autoimmune encephalomyelitis (EAE) rats. Methods Immunohistochemistry was used to detect the expression of vascular cell adhesion molecule 1 (VCAM 1) and major histocompatibility complex (MHC) class II molecules on cerebrovascular endothelial cells of EAE rats. The levels of serum TNF and IFN γ in rats with different severity were detected by MTT assay. Results EAE rat cerebrovascular endothelial cells induced expression of VCAM 1 and MHC class II molecules, TNF and IFN γ in serum. Conclusion The changes of the surface molecules of cerebrovascular endothelial cells under central nervous system inflammation suggest that they may play an important role in the pathogenesis of EAE
