目的研究新型光敏剂CDHS801在膀胱癌T24细胞内的亚细胞定位。方法将传代培养的膀胱癌T24细胞与CDHS801共同孵育,采用激光共聚焦显微镜为成像工具,应用线粒体荧光探针MitoTrackerREDCMXRose和MitoTrackerGreenFM来进行CDHS801的亚细胞定位分析。结果激光共聚焦显微镜下CDHS801、MitoTrackerREDCMXRose和MitoTrackerGreenFM发出的荧光都位于细胞浆内,在细胞核周围呈点状散在分布。结论CDHS801经膀胱癌T24细胞摄取后,定位于细胞浆内,主要分布线粒体内。
Objective To study the subcellular localization of a novel photosensitizer CDHS801 in bladder cancer T24 cells. Methods Subculturing bladder cancer T24 cells were incubated with CDHS801. The laser scanning confocal microscope was used as the imaging tool. Mitochondrial fluorescent probes MitoTracker REDCMXRose and MitoTracker Green FM were used to analyze the subcellular localization of CDHS801. Results The fluorescence of CDHS801, MitoTracker REDCMXRose and MitoTracker Green MF were all localized in the cytoplasm under the confocal laser scanning microscope, and scattered around the nucleus. Conclusion CDHS801 is localized in the cytoplasm after it is taken up by bladder cancer T24 cells and mainly distributed in mitochondria.
