目的建立大鼠血浆中胃能方有效组分中柚皮苷、新橙皮苷和柴胡皂苷b2的HPLC血药浓度测定方法。方法血浆样品用甲醇沉淀蛋白,高效液相色谱法(HPLC)测定血药浓度。色谱柱为Agilent Eclipse XDB-C18(4.6 mm×250 mm,5μm);流动相:乙腈-0.1%磷酸水,流速:1 mL/min,检测波长:280 nm和254 nm。结果各成分的最低检测浓度在0.067~0.272 mg/L,平均回收率均在85%以上,日内日间精密度及稳定性的RSD均小于10%。结论该方法简便、准确,可用于胃能方有效组分中柚皮苷等3种成分的血药浓度定量分析。
Objective To establish a HPLC method for the determination of naringin, neohesperidin and saikosaponin b2 in the plasma of rats. Methods Plasma samples were precipitated with methanol and analyzed by HPLC. The column was Agilent Eclipse XDB-C18 (4.6 mm × 250 mm, 5 μm). The mobile phase was acetonitrile-0.1% phosphoric acid water and the flow rate was 1 mL / min. The detection wavelength was 280 nm and 254 nm. Results The minimum detectable concentration of each component ranged from 0.067 to 0.272 mg / L with the average recoveries above 85%. The RSDs of intra-day and intra-day precision and stability were less than 10%. Conclusion The method is simple and accurate and can be used for the quantitative analysis of plasma concentrations of three components of naringin in Weifeng recipe.
