本实验以杨树几个代表种及杨树杂交组合‘美杨×小叶杨’的正反交杂种和亲本,柳树杂交组合‘垂柳×爆竹柳’杂种及其亲本为材料,利用十二烷基磺酸钠(SDS)-聚丙烯酰胺凝胶电泳(PAGE)方法,比较分析了叶绿体类囊体膜蛋白表征,及其在亲子代之间的遗传表现和雌雄性别间的差异。结果表明,杨树叶绿体类囊体膜蛋白质的SDS-PAGE,可显示出40多条带,不同种间,膜蛋白成分存在差异。根据差异的表现,可以将供试的杨属的几个代表种划分为三组。此外,杨树雌雄性别间膜蛋白组成有差异,但不同代表种表现各不相同,没有得到一定的共同规律。小叶杨雄株比雌株多出一条稳定的分子量为63kd多肽带,可以作为小叶杨的雄性生化标志。在杨树杂交组合中,分子量为44kd的多肽带表现为偏母遗传。柳树杂种形态性状分离与其膜蛋白组成成份分离相吻合,说明细胞质对树木杂种性状的遗传具有一定的相关作用。杂种出现特异多肽带,说明膜蛋白表征可以作为一个标记,应用于杂种分析和鉴定中。
In this experiment, the hybrids of ’Yangmei × Populus simon × Populus × Populus × Populus × Populus × Populus × Euphratica × Populus × ratoon’ hybrids and several hybrids of poplar and hybrids of weeping willow × poplar × hybrids and their parents were used as materials, (SDS) -polyacrylamide gel electrophoresis (PAGE) was used to analyze the characteristics of membrane proteins of chloroplast thylakoid and to analyze their differences in genetic expression between male and female and between male and female. The results showed that there were more than 40 bands in the membrane of poplar chloroplast thylakoid membrane protein, different from each other, the membrane protein components were different. According to the performance of the differences, several representative Populus species can be divided into three groups. In addition, poplar male-female membrane protein composition differences, but different representatives of different performance, did not get a certain common law. Populus xiongxiong more than the female plant a stable molecular weight of 63kd peptide band, can be used as male poplar male biochemical markers. In the poplar hybrid combination, the 44 kd molecular weight polypeptide band showed a partial mother heredity. The segregation of hybrid morphological traits of willow trees is consistent with the separation of their membrane protein components, indicating that cytoplasm has a certain correlation with the inheritance of tree hybrid traits. Hybrids with specific polypeptide bands, indicating that membrane protein characterization can be used as a marker, used in hybrid analysis and identification.
