[目的]研究葫芦素B对卵巢癌细胞SKOV3的生长抑制作用。[方法]利用MTT法检测不同浓度葫芦素B对卵巢癌细胞的增殖抑制作用;流式细胞术检测细胞周期和凋亡情况;荧光定量PCR法测定STAT3 m RNA表达水平。[结果]葫芦素B对SKOV3细胞的增殖具有抑制作用,并呈现时间和剂量依赖性(P<0.05);随葫芦素浓度的增加,出现明显的G2/M期细胞阻滞,细胞凋亡率也随之增加(P<0.05);STAT3 m RNA表达量明显下降(P<0.05)。[结论]葫芦素B抑制STAT3的表达,引起细胞周期阻滞和诱导细胞凋亡,抑制卵巢癌细胞的生长。
[Objective] To study the inhibitory effect of cucurbitacin B on the growth of ovarian cancer cell line SKOV3. [Method] MTT assay was used to detect the proliferation of ovarian cancer cells with different concentrations of cucurbitacin B. The cell cycle and apoptosis were detected by flow cytometry. The expression of STAT3 mRNA was detected by real-time PCR. [Result] Cucurbitacin B had an inhibitory effect on the proliferation of SKOV3 cells in a time and dose-dependent manner (P <0.05). With the increase of cucurbitacin concentration, significant G2 / M phase cell arrest and apoptotic rate Also increased (P <0.05). The STAT3 m RNA expression decreased significantly (P <0.05). [Conclusion] Cucurbitacin B inhibits the expression of STAT3, induces cell cycle arrest, induces cell apoptosis and inhibits the growth of ovarian cancer cells.
