背景:社会环境压力可引起交感神经-肾上腺髓质系统兴奋,释放大量的肾上腺素和去甲肾上腺素加速促进骨髓造血干细胞向淋巴细胞和浆细胞分化,粒细胞数降低。这种现象可以帮助机体缓冲和适应内外环境刺激,保护一些重要器官不受损伤。目的:观察社会环境压力对中国树鼩血液学和骨髓象指标的影响及利血平的干预作用。设计:性别配对,对照试验。材料:选用健康成年中国树鼩30只,雌雄各半,体质量120~140g。自制实验筒:PVC管长16cm,内径6cm,一端用玻璃封闭,另一端有插板,筒上有孔用于通气孔。SHZ-82型气浴恒温振荡器(常州医疗器件厂生产)。方法:实验于2003-03/10在甘肃政法学院公安分院实验中心实验室完成。①将中国树鼩按性别配对原则分为5组,每组6只:对照组:正常饲养;利血平+振动组:实验前4h肌肉注射肾上腺素能神经元阻断性抗高血压药利血平0.05mg/kg,振动140次/min;低、中、高频率振动组:分别振动70,140和210次/min。除对照组外,其他各组动物分别装入实验筒内放置5min,再放入振动器内按各自频率振动2min。②用Duke法和毛细玻管法测出血时间、凝血时间;血沉管法测定血沉。用全自动721型生化仪测定血细胞和骨髓象各指标。③计量资料差异比较采用单因素方差分析。主要观察指标:各组中国树鼩血液学和骨髓象指标比较。结果:中国树鼩30只均进入结果分析。①血液学指标:中频率振动组动物出血时间比对照组延长了104.8%(P<0.01),凝血时间和血沉分别缩短了37.3%和7.5%(P<0.01),红细胞数、网织红血球数、血红蛋白含量、血小板和白细胞数增加了17.5%,50.9%,11.8%,10.3%和13.7%(P<0.05~0.01);利血平+振动组与对照组比较,差异不明显(P>0.05);各频率振动组间差异也不明显(P<0.05)。②骨髓象指标:中频率振动组动物骨髓粒细胞数比对照组降低了31.5%(P<0.01),红细胞、淋巴细胞、单核细胞、浆细胞、巨核细胞和有丝分裂细胞增加了25.7%,77.6%,213.3%,65.8%,100%和161.3%(P<0.01);利血平+振动组与对照组相近(P>0.05);各频率振动组间差异也不明显(P>0.05)。结论:制动并给予不同频率振动可使中国树鼩血细胞数目明显增加,骨髓粒细胞数降低,这种变化可被利血平干预,且不随振动频率改变而变化。
BACKGROUND: Social and environmental stress can cause sympathetic - adrenal system excitement, releasing large amounts of epinephrine and norepinephrine to accelerate the differentiation of bone marrow hematopoietic stem cells into lymphocytes and plasma cells, reducing the number of granulocytes. This phenomenon can help the body buffer and adapt to internal and external environment to stimulate, protect some important organs from damage. Objective: To observe the influence of social environment pressure on the hematology and bone marrow index of Chinese tree gall and the effect of reserpine. Design: Gender matching, controlled trial. Material: Choose healthy adult tree trunks 30, male and female, body weight 120 ~ 140g. Self-made experimental tube: PVC pipe length 16cm, diameter 6cm, closed at one end with glass, the other end of the board, a hole on the cylinder for the vent. SHZ-82-type gas bath temperature oscillator (Changzhou Medical Devices Factory production). Methods: The experiment was performed in the Laboratory of Public Security Branch Laboratory of Gansu Political Science and Law Institute from March to March 2003. ① The tree shrews were divided into 5 groups according to the principle of gender pairing, 6 in each group: control group: normal feeding; reserpine + sham group: intramuscular injection of adrenergic neurons blocking antihypertensive drugs Blood 0.05mg / kg, vibration 140 beats / min; low, medium and high frequency vibration group: vibration 70,140 and 210 times / min. In addition to the control group, the other groups of animals were placed in the experimental tube placed 5min, then placed in a vibrator at their own frequency vibration 2min. ② Duke method and capillary tube method to determine the blood time, clotting time; ESR ESR. Using automatic 721 biochemical analyzer blood cells and bone marrow as the indicators. ③ measurement data differences using one-way analysis of variance. MAIN OUTCOME MEASURES: Hematology and bone marrow index of tree shrews in each group were compared. Results: Thirteen Chinese shrews were all included in the result analysis. ① Hematological parameters: The frequency of hemorrhage in the medium frequency vibration group was prolonged by 104.8% (P <0.01), and the clotting time and erythrocyte sedimentation time were 37.3% and 7.5% (P <0.01), respectively. The numbers of red blood cells, (P <0.05 ~ 0.01). The level of hemoglobin, platelet and leucocyte increased by 17.5%, 50.9%, 11.8%, 10.3% and 13.7% ). There was no significant difference between each frequency vibration group (P <0.05). (2) Bone marrow index: Compared with the control group, the number of myeloid granulocytes in the medium-frequency vibration group decreased by 31.5% (P <0.01), the increase of erythrocytes, lymphocytes, monocytes, plasma cells, megakaryocytes and mitotic cells by 25.7% and 77.6 %, 213.3%, 65.8%, 100% and 161.3%, respectively (P <0.01). Reserpine + vibration group was similar to the control group (P> 0.05). CONCLUSION: Braking and vibration at different frequencies can significantly increase the number of blood cells in tree shrews and decrease the number of myeloid granulocytes. This change may be influenced by reserpine without changing with the frequency of vibration.
