目的探讨人乳腺球蛋白(hMAM)、血管内皮生长因子-C(VEGF-C)及其受体-3(VEGFR-3)在乳腺癌组织中表达,分析其表达与乳腺癌生物学特性及预后的关系。方法采用组织微阵列技术,通过免疫组化法检测116例乳腺癌组织、44例癌旁正常组织中hMAM、VEGF-C及VEGFR-3的表达。结果 (1)乳腺癌组织中hMAM、VEGF-C及VEGFR-3的表达率分别为59.48%(69/116),50.86%(59/116)及61.20%(71/116),均高于癌旁正常组织中的表达率(分别为0.00%、9.09%及18.18%,P均<0.01)。(2)hMAM、VEGF-C及VEGFR-3的表达与腋淋巴结转移呈正相关(P均<0.05),hMAM和VEGFR-3的表达与组织学分级呈正相关(P均<0.05)。(3)采用Spearman等级相关分析结果显示,hMAM、VEGF-C及VEGFR-3在乳腺癌中的表达两两之间均呈正相关,相关系数分别为0.278、0.280及0.244(P均<0.01)。(4)Kaplan-Meier生存分析结果表明:hMAM、VEGF-C及VEGFR-3阴性表达者无病生存率及5年总生存率均高于三者阳性表达(Log-rank,P均<0.01)。结论 hMAM、VEGF-C及VEGFR-3在乳腺癌组织中存在高表达,其高表达与乳腺癌的发生、侵袭转移和预后有关,且hMAM可能与乳腺癌微淋巴管生成有关。
Objective To investigate the expression of human mammary gland protein (hMAM), vascular endothelial growth factor-C (VEGF-C) and its receptor-3 (VEGFR-3) in breast cancer tissues and their biological characteristics and prognosis Relationship. Methods Tissue microarray was used to detect the expression of hMAM, VEGF-C and VEGFR-3 in 116 cases of breast cancer tissues and 44 cases of adjacent normal tissues by immunohistochemistry. Results (1) The positive rates of hMAM, VEGF-C and VEGFR-3 in breast cancer tissues were 59.48% (69/116), 50.86% (59/116) and 61.20% (71/116) Adjacent normal tissues (0.00%, 9.09% and 18.18%, P <0.01 respectively). (2) There was a positive correlation between hMAM, VEGF-C and VEGFR-3 expression and axillary lymph node metastasis (all P <0.05). The expression of hMAM and VEGFR-3 was positively correlated with histological grade (all P <0.05). (3) Spearman rank correlation analysis showed that there was a positive correlation between the expression of hMAM, VEGF-C and VEGFR-3 in breast cancer, the correlation coefficients were 0.278,0.280 and 0.244 respectively (all P <0.01). (4) Kaplan-Meier survival analysis showed that the disease-free survival rate and 5-year overall survival rate of hMAM, VEGF-C and VEGFR-3 negative patients were all higher than those of the three positive patients . Conclusion The expression of hMAM, VEGF-C and VEGFR-3 in breast cancer tissues is high. The high expression of hMAM, VEGF-C and VEGFR-3 is related to the occurrence, invasion, metastasis and prognosis of breast cancer, and hMAM may be related to the lymphangiogenesis of breast cancer.
