【目的】探讨铁皮石斛种子萌发、芽增殖和生根的适宜培养基,为铁皮石斛组织快繁技术提供依据。【方法】以铁皮石斛种子为外植体进行培养,研究6-BA(0-3.0mg/L)、NAA(0.1-0.3mg/L)、香蕉泥(15%~25%)和马铃薯泥(15%~25%)对铁皮石斛种子萌发、芽增殖和生根壮苗的影响。【结果】适宜种子萌发的较好培养基为MS+20%马铃薯泥,芽增殖最适培养基为MS+2.0mg/L6-BA,增殖率达282.7%;最适生根培养基为MS+15%香蕉泥,生根率达100%。【结论】经马铃薯泥、香蕉泥培养的铁皮石斛组培苗,生长健壮,能为生产提供大量的种苗,因而此类培养基可用于大规模生产铁皮石斛组培苗。
【Objective】 The purpose of this study was to investigate the suitable medium for the germination, bud proliferation and rooting of Dendrobium candidum, providing the basis for rapid propagation of Dendrobium officinale. 【Method】 The seeds of Dendrobium candidium were used as explants to study the effects of 6-BA (0-3.0mg / L), NAA (0.1-0.3mg / L), banana puree (15% -25% 15% ~ 25%) on the seed germination, bud proliferation and rooting strong seedling of Dendrobium officinale. 【Result】 The optimum culture medium for seed germination was MS + 20% mashed potatoes. The optimum medium for bud proliferation was MS + 2.0 mg / L 6-BA with a proliferation rate of 282.7%. The optimum medium for rooting was MS + 15 % Banana puree, rooting rate of 100%. 【Conclusion】 The tissue culture seedlings of Dendrobium candidum cultured in mashed potatoes and banana puree can grow robustly and provide a large number of seedlings for production. Therefore, such culture medium can be used for large-scale production of Dendrobium candidum tissue culture seedlings.
