目的探讨益气温阳活血方对慢性心力衰竭心室重塑大鼠β3-肾上腺素能受体(β3-AR)、血清Ⅰ型前胶原羧基端肽(PⅠCP)和Ⅲ型前胶原氨基端肽(PⅢNP)的影响。方法通过异丙肾上腺素(ISO)诱导慢性心力衰竭动物模型。计算左、右心室心肌的心脏指数(LVMI、RVMI),应用酶联免疫吸附(ELISA)法检测血清PⅠCP和PⅢNP含量,以逆转录聚合酶链反应(RT-PCR)法以及Western印迹法检测β3-AR mRNA及蛋白质表达水平。结果与正常组比较,模型组LVMI〔(2.987±0.318 vs 2.350±0.468)mg/g〕、血清PⅠCP〔(16.357±3.650 vs 12.843±3.124)pg/ml〕、PⅢNP〔(64.984±17.477 vs 21.245±5.354)pg/ml〕含量及β3-AR mRNA(0.888±0.064 vs 0.743±0.086)与蛋白质(4.413±0.729vs 2.312±0.394)的表达水平明显升高(均P<0.05);益气温阳活血方能够明显降低慢性心力衰竭大鼠LVMI,减少PⅠCP和PⅢNP含量,抑制β3-AR mRNA与蛋白质表达水平的升高,与正常组比较〔(2.557±0.379 vs 2.350±0.468)mg/g〕、血清PⅠCP〔(13.111±2.758 vs 12.843±3.124)pg/ml〕和PⅢNP〔(25.967±9.659 vs 21.245±5.354)pg/ml〕含量及β3-AR mRNA(0.781±0.067 vs 0.743±0.086)与蛋白质(2.484±0.642 vs 2.312±0.394),均无显著性差异(P>0.05)。结论益气温阳活血方可能通过抑制β3-AR的表达减少PⅠCP和PⅢNP含量逆转心室重塑,改善心脏功能。
Objective To investigate the effect of Yiqi Wenyang Huoxue Decoction on β3-adrenergic receptor (β3-AR), serum type Ⅰ procollagen carboxyterminal peptide (PⅠCP) and type Ⅲ procollagen aminoterminal peptide (PⅢNP) in ventricular remodeling rats with chronic heart failure )Impact. Methods Animal models of chronic heart failure were induced by isoproterenol (ISO). The left and right ventricular myocardium cardiac index (LVMI, RVMI) were calculated. The contents of PⅠCP and PⅢNP in serum were detected by enzyme-linked immunosorbent assay (ELISA) -AR mRNA and protein expression levels. Results Compared with the normal group, LVMI 〔(2.987 ± 0.318 vs 2.350 ± 0.468) mg / g〕, PⅠCP [(16.357 ± 3.650 vs 12.843 ± 3.124) pg / ml〕, PⅢNP 〔(64.984 ± 17.477 vs 21.245 ± 5.354) pg / ml〕 and the expression of β3-AR mRNA (0.888 ± 0.064 vs 0.743 ± 0.086) and protein (4.413 ± 0.729 vs 2.312 ± 0.394) (all P <0.05) (2.557 ± 0.379 vs 2.350 ± 0.468) mg / g, PⅠCP, PⅠCP and PⅠCP in rats with chronic heart failure were significantly lower than those in normal rats (P <0.05) (13.111 ± 2.758 vs 12.843 ± 3.124 pg / ml) and PⅢNP (25.967 ± 9.659 vs 21.245 ± 5.354 pg / ml), and the levels of β3-AR mRNA (0.781 ± 0.067 vs 0.743 ± 0.086 and 2.484 ± 0.642 vs 2.312 ± 0.394), no significant difference (P> 0.05). Conclusion Yiqi Yangyanghuoxue Prescription can reverse ventricular remodeling and improve cardiac function by decreasing the expression of PⅠCP and PⅢNP by inhibiting the expression of β3-AR.
