目的:观察淡豆豉异黄酮对血管紧张素Ⅱ(Angiotensin Ⅱ,Ang Ⅱ)诱导大鼠血管平滑肌细胞(Vascular Smooth MuscleCell,VSMC)细胞周期的影响,探讨其抑制增殖的可能机制。方法:原代培养大鼠VSMC,建立AngⅡ诱导VSMC增殖模型,MTT法观察淡豆豉异黄酮对Ang Ⅱ诱导VSMC增殖的干预作用,流式细胞仪观察淡豆豉异黄酮干预细胞后细胞周期的变化。结果:淡豆豉异黄酮50μg/L预孵育12、24 h,100μg/L预孵育1、4、12、24 h,200μg/L预孵育1、4、24 h,500μg/L预孵育4 h可显著抑制AngⅡ诱导的VSMC增殖;50、100、200μg/L作用24 h抑制增殖作用最为明显,并可显著增加VSMC G0/G1期比例,降低S期比例。结论:淡豆豉异黄酮抑制Ang Ⅱ诱导的VSMC增殖作用机制可能与阻止细胞由G0/G1期进入S期,进行VSMC细胞周期的调控有关。
Objective: To observe the effect of isoflavone on the cell cycle of rat vascular smooth muscle cells (VSMC) induced by Angiotensin Ⅱ (Ang Ⅱ), and to explore its possible mechanism of inhibiting proliferation. Methods: VSMCs were cultured in primary culture to establish the model of VSMC proliferation induced by AngⅡ. The effects of isoflavones on the proliferation of VSMC induced by Ang Ⅱ were observed by MTT assay. The changes of cell cycle were observed by flow cytometry. Results: The pre-incubation of isoflavones 50 mg / L was 12,24 h, preincubation at 100 μg / L for 1, 4, 12, 24 h, preincubation at 200 μg / L for 1, 4, Significantly inhibited the proliferation of VSMC induced by AngⅡ. The effects of 50,100,200μg / L for 24 h were most obvious, and the proportion of G0 / G1 phase in VSMC was significantly increased and the proportion of S phase was decreased. CONCLUSION: The mechanism by which isoflavone inhibits the proliferation of VSMC induced by Ang Ⅱ may be related to preventing cells from entering the S phase through G0 / G1 phase and regulating the cell cycle of VSMC.
