目的探讨野生型p53基因及其表达产物p53蛋白在恶性肿瘤基因治疗中的作用。方法应用腺病毒为载体将野生型p53基因转入高、低转移的肺癌细胞系Anip973、AGZY83-a,并且对各组转染细胞进行生长曲线、原位末端标记、Western-blotting等技术检测分析。结果野生型p53蛋白的过表达对上述肺腺癌细胞系均呈现出较强的生长抑制作用,野生型p53蛋白的过表达对高转移肺癌细胞系Anip973的抑制作用明显高于低转移细胞系AGZY83-a。结论外源性野生型p53基因的过表达,可以有效抑制上述肿瘤细胞的增殖,同时对高转移肺腺癌细胞系的抑制作用更加显著。
Objective To investigate the role of wild type p53 gene and p53 protein in gene therapy of malignant tumors. Methods Wild type p53 gene was transfected into high and low metastatic lung cancer cell lines Anip973 and AGZY83-a using adenovirus as carrier, and the growth curve, in situ end-labeling and Western-blotting were used to detect the expression of wild-type p53 gene. . Results The overexpression of wild-type p53 protein showed a strong growth inhibitory effect on the above-mentioned lung adenocarcinoma cell lines. The overexpression of wild-type p53 protein was significantly higher than that of low metastatic cell line AGZY83 -a. Conclusion Overexpression of exogenous wild-type p53 gene can effectively inhibit the proliferation of the above-mentioned tumor cells and inhibit the proliferation of the highly-metastatic lung adenocarcinoma cell line more significantly.
