目的本试验以脱毒SH40和用60Co-γ射线辐射诱变的SH40试管苗为试材,研究了60Co对SH40叶片再生和植株形态的影响.方法运用组织培养技术比较辐射前后SH40的叶片再生率,观察继代苗的植株形态并且测定叶绿素含量.结果在相同的培养条件下,当TDZ浓度为1.5 mg/L和2.0 mg/L时,经辐射试管苗的叶片再生率以及平均出芽是都有所降低,并且叶裂加深、偏叶、皱缩叶、花斑叶、褪绿叶、漏斗叶、狭长叶、一柄两叶所占的比例明显增加;辐射使幼苗的生长受到抑制,增值系数和叶绿素含量明显减少.结论辐射诱变抑制了SH40试管苗的叶片再生,使叶片的外形发生了较大的变化,畸形叶所占比例增大,植株的苗高、增值系数和叶绿素含量降低.
Objective To study the effects of 60Co on SH40 leaf regeneration and plant morphology using detoxified SH40 and SH40 strain induced by 60Co-γradiation.METHODS Tissue culture techniques were used to compare leaf regeneration rate , The plant morphology of the subculture was observed and the content of chlorophyll was measured.Results Under the same culture conditions, when the TDZ concentration was 1.5 mg / L and 2.0 mg / L, the leaf regeneration rate and average budding of the irradiated plantlets were Decreased, and the deepening of the leaf cleft, the proportion of leaves, folds, leaf spotted leaves, chlorotic leaves, funnel leaves, narrow leaves, a handle and two leaves significantly increased; radiation to seedling growth was inhibited, The content of chlorophyll decreased significantly.Conclusion Radiation induced mutagenesis inhibited the leaf regeneration of SH40 plantlets, which led to the bigger change of leaf shape, the increased proportion of deformed leaf, and the plant height, value added coefficient and chlorophyll content decreased.
