食用菌深层培养技术用于制作原种,栽培种已显示出很多优越性。本项研究旨在探讨液体培养下稀土元素对猴头菇的生理效应及栽培效果。现将试验结果报告如下: 一、材料与方法 (一)培养基 ①斜面培养基:以PDA为基础培养基,添加磷酸氢二钾1g,硫酸镁0.3g,酵母膏5g,用柠檬酸调pH5.5。②液体培养基:上述配方去琼脂。③固体培养基:棉子壳86%,麸皮10%,石膏粉2%,过磷酸钙2%。 (二)试验方法 取猴头菇菌种斜面接种在液体培养基上,振荡培养10天,无菌过滤得菌丝体,用等体积生理盐水稀释摇匀得标准接种液。液体培养基添加不同浓度的稀土。用250ml三角瓶,盛稀土培养液80ml,灭菌后均一接种,振荡培养并定期检测菌丝干重。以培养12天的猴头液体菌种制备小麦颗粒菌种,在无菌条件下接入灭菌棉子壳培养基料袋两头。常规培养与管理,跟踪观测各种性状表现。
Edible fungi deep culture technology for the production of original species, cultivars have shown many advantages. This study aimed to investigate the physiological effects of rare earth elements on Hericium erinaceus under liquid culture and its cultivation effect. Now the test results are reported as follows: I. Materials and Methods (a) medium ① slant medium: PDA-based medium, add dipotassium hydrogen phosphate 1g, magnesium sulfate 0.3g, yeast extract 5g, with citric acid pH5 .5. ② liquid medium: the above formula to agar. ③ solid medium: Cotton Seed Shell 86%, bran 10%, gypsum powder 2%, superphosphate 2%. (B) Test Method Take Hericium Mushroom slant inoculated on liquid medium, shake culture for 10 days, mycelium was sterile filtered, diluted with an equal volume of saline diluted standard inoculum. Liquid medium to add different concentrations of rare earth. With 250ml flask, Sheng Rare Earth culture medium 80ml, uniformly inoculated after sterilization, shake culture and regular detection of mycelial dry weight. To cultivate 12 days of Hericium liquid bacterial species of wheat particles were prepared, access to sterilized cottonseed shell under both bacterial culture media bags. Routine training and management, tracking and observing the performance of various traits.
