目的探讨安多霖对微波辐射致大鼠生殖远后损伤是否有保护作用及有效剂量。方法二级Wistar雄性大鼠80只,随机分为4组,即正常对照组、辐射对照组、1.5 g安多霖组、3.0 g安多霖组,采用30 mW/cm2微波辐射大鼠,于辐射前14 d开始灌胃给药,每天1次,连续14 d,安多霖组给药量(给药浓度)为1.5 g/(kg·d)和3 g/(kg·d);正常对照组和辐射对照组给予等比体积的蒸馏水。各组大鼠分别于辐射(停药)后1 m、3 m用1%戊巴比妥钠(30 mg/kg)经腹腔注射麻醉处死大鼠,利用精子动态分析系统分析精子活动度,HE染色观测并计数精子畸形率,光镜和电镜观察睾丸形态结构。结果 (1)30 mW/cm2微波辐射后1 m,精子活动度明显减少,D级精子比例增加,睾丸组织生精上皮疏松,偶见变性和坏死的脱落细胞,生精细胞核染色质分布不均,退行性病变,线粒体肿胀空化。(2)停药后1 m,与辐射对照组相比,1.5 g安多霖预防组,C级精子比例增加(P<0.01);3 g安多霖组精子A级、A+B级精子比例增加、C级和D级精子比例减小(P<0.01),活力参数曲线速度(VCL)、直线速度(VSL)、平均速度(VAP)、平均振幅(ALH)均明显升高(P<0.05或P<0.01);睾丸组织结构和超微结构明显改善。结论 30 mW/cm2微波辐射可致大鼠生殖功能和结构远后损伤,表现为精子的活动度下降;睾丸组织生精上皮疏松、精原细胞染色质分布不均,退行性病变。预防应用1.5 g和3 g/(kg·d)对上述损伤具有一定的保护作用,1.5 g/(kg·d)为有效剂量。
Objective To investigate whether Andorin can protect rats from long-term reproductive injury caused by microwave radiation and its effective dose. Methods Eighty Wistar male rats were randomly divided into 4 groups: normal control group, radiation control group, 1.5 g Andoline group, and 3.0 g Andolin group. Rats were exposed to 30 mW / Radix Astragali was administrated intragastrically once a day for 14 days before radiation, and the dose of amlodrine was 1.5 g / (kg · d) and 3 g / (kg · d) respectively. Normal Control group and radiation control group were given equal volume of distilled water. The rats were killed by intraperitoneal injection of 1% pentobarbital sodium (1 mg / kg) 1 m and 3 m respectively after irradiation (withdrawal), and sperm motility was analyzed by sperm dynamic analysis system. HE Dyeing observation and counting sperm deformity rate, light microscope and electron microscope testicular morphology. Results (1) At 1 m after 30 mW / cm2 microwave irradiation, the activity of sperm decreased significantly and the proportion of grade D sperm increased. The testicular epithelial cells were loose and the degenerated and necrotic exfoliated cells were occasionally observed. The distribution of chromatin in germ cell was uneven , Degenerative disease, mitochondria swelling cavitation. (2) Compared with the radiation control group, the proportion of C-level sperm in the 1.5-g Adolfian prevention group increased (P <0.01) (P <0.01), while the parameters of VCL, VSL, VAP and ALH increased significantly (P < 0.05 or P <0.01); testicular tissue structure and ultrastructure improved significantly. CONCLUSION: The 30 mW / cm2 microwave radiation can cause reproductive function and structural damage in the distal and distal parts of the rat. The activity of the sperm is decreased. The spermatogenic epithelium is loose and the chromatin is unevenly distributed in the spermatogonia. Preventive application of 1.5 g and 3 g / (kg · d) of these injuries have a protective effect, 1.5 g / (kg · d) is an effective dose.
