血卟啉衍生物介导的光动力疗法对人肺腺癌细胞杀伤效应的实验研究

目的研究血卟啉衍生物(hematoporphyrin derivative,HpD)介导的光动力疗法(photodynamic therapy,PDT)对在体和离体人肺腺癌细胞A549的杀伤作用。方法以MTT法检测HpD-PDT对A549细胞增殖抑制的量效关系,应用流式细胞术分析其对细胞凋亡的影响。进一步构建A549细胞裸鼠移植瘤动物模型,通过绘制肿瘤生长曲线观察HpD-PDT对肺癌生长的抑制作用,并应用TUNEL法检测肿瘤细胞凋亡。结果HpD-PDT作用后,A549细胞增殖受抑,凋亡增加,显著高于对照组(P<0.05),且其抑制效率依赖于HpD浓度及激光剂量;裸鼠移植瘤动物模型肿瘤生长受抑、凋亡增加,也与对照组存在显著性差异(P<0.01)。结论HpD-PDT可有效杀伤肺腺癌细胞,其作用机制可能与抑制肿瘤细胞增殖、诱导细胞凋亡有关。 Objective To study the killing effect of hematoporphyrin derivative (HpD) -mediated photodynamic therapy (PDT) on human lung adenocarcinoma A549 cells in vitro and in vivo. Methods MTT assay was used to detect the dose-response relationship of HpD-PDT on A549 cell proliferation. The effect of HpD-PDT on apoptosis was analyzed by flow cytometry. To further establish the animal model of A549 cell xenograft in nude mice, observe the inhibitory effect of HpD-PDT on the growth of lung cancer by drawing the tumor growth curve, and detect the apoptosis of tumor cells by TUNEL method. Results After HpD-PDT treatment, the proliferation of A549 cells was inhibited and the apoptosis increased, which was significantly higher than that of the control group (P <0.05). The inhibitory efficiency of HpD-PDT was dependent on the HpD concentration and laser dose. , Apoptosis increased, but also with the control group there was significant difference (P <0.01). Conclusion HpD-PDT can effectively kill lung adenocarcinoma cells and its mechanism may be related to the inhibition of tumor cell proliferation and induction of apoptosis.
