目的 :观察胃喜康对胃溃疡大鼠胃黏膜表皮生长因子 (EGF)及受体 (EGFR)的影响 ,探讨该药抗胃溃疡的可能机制。方法 :采用乙酸注射至大鼠浆膜下形成胃溃疡模型 ,用放射免疫法检测胃黏膜组织EGF含量 ,用原位杂交的方法检测胃黏膜组织EGFRmRNA的表达。结果 :胃喜康组溃疡指数明显低于模型组 (P <0 .0 5 ) ,而与雷尼替丁组无差异 (P >0 .0 5 ) ,胃喜康组及雷尼替丁组黏膜组织的EGF的含量显著高于模型组 (P <0 .0 1) ;模型组胃黏膜组织EGFRmRNA平均灰度值明显低于对照组 (P <0 .0 5 ) ,胃喜康组及雷尼替丁组EGFRmRNA平均灰度值显著低于模型组 (P <0 .0 5 ,P <0 .0 1) ,胃喜康组比雷尼替丁组EGFRmRNA平均灰度值更低 (P <0 .0 5 )。结论 :胃喜康的抗溃疡作用可能与激活EGF/EGFR系统 ,促进胃黏膜上皮细胞增生、组织修复 ,抑制胃酸有关。
Objective: To observe the effect of Weixikang on epidermal growth factor (EGF) and receptor (EGFR) in gastric mucosa of rats with gastric ulcer, and to explore the possible mechanism of this drug against gastric ulcer. METHODS: A gastric ulcer model was established by subcutaneous injection of acetic acid in rats. EGF content in gastric mucosa was detected by radioimmunoassay. The expression of EGFR mRNA in gastric mucosa was detected by in situ hybridization. Results: The ulcer index of Weixikang group was significantly lower than that of model group (P < 0.05), but it was not different from ranitidine group (P > 0.05), Weixikang group and ranitidine group. The EGF content in mucosal tissues was significantly higher than that in the model group (P < 0.01); the average gray value of EGFR mRNA in the gastric mucosal tissue in the model group was significantly lower than that in the control group (P <0.05). Weixikang group and Lei The average gray value of EGFR mRNA in the nitinidine group was significantly lower than that in the model group (P <0.05, P <0.01), and the mean gray value of EGFR mRNA was lower in the Weixikang group than in the ranitidine group (P < 0 .0 5 ). Conclusion: The anti-ulcer effect of Weixikang may be related to the activation of EGF/EGFR system, promotion of gastric mucosal epithelial cell proliferation, tissue repair, and inhibition of gastric acid.
