目的了解缺氧缺血性脑损伤(HIBD)对松果体芳香烷基胺-N-乙酰基转移酶(AANAT)mRNA表达和血浆褪黑素(MLT)含量的影响,探讨松果体功能改变在HIBD中的意义。方法选取7日龄SD大鼠60只随机分成HIBD模型组和假手术组。HIBD模型组按改良Levine法建立,然后用半定量逆转录聚合酶链反应(RT-PCR)和放免技术分别测定并比较两组缺氧缺血后0 h、6 h、12 h、24 h、48 h松果体中AANAT mRNA的表达水平及血浆MLT的浓度。结果 (1)松果体中AANATmRNA的表达在HIBD后6 h、12 h、24 h低于对应假手术组(P<0.05或<0.01);假手术组各时点之间AANAT mRNA的表达水平无明显变化(P>0.05)。(2)血浆MLT浓度在HIBD后12 h、24 h均低于对应假手术组(P<0.01);假手术组各时点之间MLT浓度均无明显变化(P>0.05)。结论 HIBD早期松果体合成褪黑素的功能下降及内源性MLT保护作用的降低可能参与早期HIBD的发病。
Objective To investigate the effect of hypoxic-ischemic brain damage (HIBD) on the expression of AANAT mRNA and the content of melatonin (MLT) in pineal gland, and to explore the changes of the function of pineal gland Meaning in HIBD. Methods Sixty SD rats of 7 days old were randomly divided into HIBD model group and sham operation group. The HIBD model group was established by modified Levine method. Then the mRNA and protein expression of HIBD was determined by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) and radioimmunoassay (RIA), and compared between two groups at 0 h, 6 h, 12 h, 24 h, The expression level of AANAT mRNA and the concentration of plasma MLT in pineal gland at 48 h. Results (1) The expression of AANAT mRNA in pineal gland was lower than that in sham group at 6 h, 12 h and 24 h after HIBD (P <0.05 or <0.01). The expression of AANAT mRNA No significant change (P> 0.05). (2) The concentration of plasma MLT was lower at 12 h and 24 h after HIBD than that of corresponding sham operation group (P <0.01). There was no significant change in MLT concentration between sham operation group and control group (P> 0.05). Conclusions The decrease of the function of melatonin synthesis and the decrease of endogenous MLT protection in early stage of HIBD may be involved in the pathogenesis of early HIBD.
