目的利用实时定量PCR(RQ-PCR)检测乳腺癌患者内皮素-2 mRNA(EDN-2)的表达情况。方法选取EDN-2 mRNA特异性序列作为检测靶基因设计引物,用SYBR Green$建立20μl实时定量反应体系,对160份乳腺癌组织和148例正常乳腺组织进行RQ-PCR检测。结果与正常乳腺组织相比,EDN-2 mRNA在乳腺癌组织中表达含量明显升高,与正常乳腺组织之间存在显著性差异(P<0.01)。EDN-2 mRNA表达含量的增加与患者的年龄和肿瘤的组织类型无关(P>0.05),只和肿瘤的大小(P<0.05)、是否有淋巴结转移(P<0.01)以及肿瘤病理级别(P<0.01)有关。结论 EDN-2 mRNA在乳腺癌中高度表达,且含量随着肿瘤越大,病理级别越高,淋巴结转移数目越多,其表达量也越高。EDN-2 mRNA的检测可作为乳腺癌的辅助检查,其含量的变化也可作为病情的发展和预后的观察指标,为乳腺癌的诊断与治疗提供新的途径,该检测项目宜在临床工作中得到开展。
Objective To detect the expression of endothelin-2 mRNA (EDN-2) in breast cancer patients by real-time quantitative PCR (RQ-PCR). Methods The specific primers of EDN-2 mRNA were selected as the primers for target gene detection. Real-time quantitative PCR reaction system was established with SYBR Green $ to detect RQ-PCR in 160 breast cancer tissues and 148 normal breast tissues. Results Compared with normal breast tissue, the expression of EDN-2 mRNA in breast cancer tissues was significantly higher than that in normal breast tissues (P <0.01). The increase of EDN-2 mRNA expression was not associated with the age and tumor type (P> 0.05), but only with tumor size (P <0.05), lymph node metastasis (P <0.01) and tumor pathological grade <0.01). Conclusion The expression of EDN-2 mRNA is highly expressed in breast cancer. The content of EDN-2 mRNA increases with the tumor size and pathological grade, and the more the number of lymph node metastasis, the higher the expression of EDN-2 mRNA. Detection of EDN-2 mRNA can be used as an auxiliary examination for breast cancer. The content of EDN-2 mRNA may also be used as an indicator of disease progression and prognosis, providing a new approach for the diagnosis and treatment of breast cancer.
