将携带有人粒细胞-巨噬细胞集落刺激因子(GM-CSF)基因的重组腺病毒载体转染BALB/C小鼠肝癌细胞株(H22),体外经60Cγ射线灭活后制成瘤苗,用以治疗行瘤小鼠。应用流式细胞仪(FCM)测定荷瘤小鼠外周血T淋巴细胞亚群的变化,并观察荷瘤小鼠生存期。结果发现,GM-CSF基因转染瘤苗能显著提高小鼠外周血CD8+T淋巴细胞亚群的含量,荷瘤小鼠生存期显著延长。结果表明:GM-CSF基因转染瘤苗能诱导出有效的抗肿瘤免疫反应,提示应用该疗法进行肿瘤基因治疗的可行性。
The recombinant adenoviral vector carrying human granulocyte-macrophage colony-stimulating factor (GM-CSF) gene was transfected into a BALB/C mouse hepatoma cell line (H22) and inactivated in vitro by 60C gamma irradiation to produce tumor vaccines. To treat tumors in mice. Flow cytometry (FCM) was used to measure the changes of peripheral T lymphocyte subsets in tumor-bearing mice and the survival of tumor-bearing mice was observed. The results showed that the GM-CSF gene transfected tumor vaccine can significantly increase the content of CD8+ T lymphocyte subsets in mice peripheral blood, and the survival period of tumor-bearing mice was significantly prolonged. The results showed that: GM-CSF gene transfected tumor vaccine can induce an effective anti-tumor immune response, suggesting the feasibility of this therapy for tumor gene therapy.
